Elimination of DNA in Namalva Interferon Preparation by Antibody Affinity Chromatography

In this report,we present the study of determination of DNA in dif-ferent Namalva interferon preparations by a microassay method usingethidium bromide.The results show that the interferon purified by anti-body affinity chromatography contains no DNA,thus indicating this pu-rified interferon preparation can be used clinically.     

Purification of Avidin with Affinity Chromatography on 2-1minobiotin-AH-Sepharose 4B

The affinity between 2-iminobiotin and avidin decreases with the decreasing ofpH.According to this property,avidin was purified in its natural state without alterationof its properties with affinity chromatography on 2-iminobiotin-AH-sepharose 4B column.The purified avidin had high activity to bind with biotin,showed one band onpolyacrylamide electrophoresis,and was much purer than the products commerciallyavailable.

Isolation of CIq in Human Serum with Anti-human CIq McAb Affinity Column Chromatography

This paper is to report a procedure for the isolation of Clq in humanserum with anti-human C1q McAb affinity column chromatography.Thisprocedure is simple and rapid,Which can be performed within 24 to 48 hours.The yield of Clq can reach 7-10 mg per 100 ml serum.The preparation isimmunoglobulin free as proved by double diffusion and immunoelectrophoresisassay and exhibits typical C1q subunit structure on SDS-PAGE.After biologicalactivity evaluation with red blood cell agglutination and latex agglutinationassay,this C1q preparation was found to be satisfactory in its activity.The anti-human C1q McAb sepharose 4B column can be used repeatedly.Itcan be stored in 4℃ for approximately a year without losing any of its absorptionactivity.     

Determination of Pyrethroids in Human Urine by Gas Chromatography

A gas chromatographic method for the determination of deltamethrin and fenvalerate in human urine is described.Both deltamethrin and fenvalerate have been analysed by gas chromatography with a electron capture detector.The least detectable concentration of both pyrethroids in urine is 0.2g per litre.The precision and accuracy of the method were within acceptable limits.The method has been used to determine the pyrethroids in urine samples obtained from packers,spraymen and acute pyrethroids poisoing patients.

Purification of Fibronectin in Human Plasma and the Preparation of Its Antiserum

The techniquc of purification of plasma fibroncctin and preparation of its antiserum is re-ported in this paper.Blood plasma was passed through an affinity column of gclatin-Sepharose 4B,fol-lowed by passing throughia column of Scphadex G200.Fibronectin with high purity was obtained.The prepare fibronectin serum.It was found that there were not only specific immune reactions be-tween the anti-human fibronectin serum and hyman fibronectins,but also cross reactions between theanti-serum and the fibronectin from other animals.The serum can be used for the study ofimmunohistochernistry of the fibronectin.

Determination of Acrylamide Metabolite,Mercapturic Acld by High Performance Liquid Chromatography

A HPLC Assay was developed to identify and measure the metabolite of acrylamide,mercapturic acid,N-Acetyl-s-(propionamid)-cysteine(APC)in urine.O-phthalaldehyde(OPA)was utilized as a precolumn derivatizing agent in the assay.This derivative was isolatedwith a good selectivity by high performance liquid chromatography(HPLC)employingreversed phase ODS columns.The quantitation of the mercapturic acid derivative wasreproducible and with a detection limit of l pmol.The average coefficient of variation for theruns carried out on the same day was approximately 4.6％ at the range of 80-160μmol·L~(-1) ofAPC,and the mean analytical recovery from urine samples was 94％. The metabolite of urine of workers exposed to acrylamide was identified as N-acetyl-s-(propionamide)-cysteine by HPLC chromatography and fluorescence scan and HPLC-Massspectra.All results were identical with the authentic synthesized compound.     

A Study on Localization of LDH-C4 on Sperm of Various Species by Monoclonal Antibody

The paper reported the results of localization of LDH-C4 on sperm of various species by immunqftuorescent test. McAbs to mouse LDH-C4 which reacted not only with mouse sperm, but also with that of other species such as rat, hamster, rabbit and human were used in this study. It is clear that LDH-C4 lacks species specificity. However, there exist different regions for localization of LDH-C4 on sperm of various species, It is suggested that gametes in various species may also differ in their LDH-C4 surface expression. The present study also showed that these McAbs to mouse LDH-C4 have different binding capacity to sperm qf varlous species. The result that LDH-C4 was mainly localized on midpiece of mouse sperm in our experiments is in discrepancy with those previously reported by other investigators.
