Ethnopharmacological relevance

Sarcopoterium spinosum (L.) sp., a common plant in the Mediterranean region, is widely used as an antidiabetic drug by Bedouin healers. However, the antidiabetic properties of Sarcopoterium spinosum had not been fully validated using scientific tools.

Aim of the study

To determine the effectiveness of Sarcopoterium spinosum extract as an antidiabetic agent in vitro and in vivo.

Materials and methods

RINm pancreatic β-cells, L6 myotubes, 3T3-L1 adipocytes and AML-12 hepatocytes were treated with an aqueous Sarcopoterium spinosum extract (0.001–10 mg/ml). The effect of the extract on specific physiological functions, including insulin secretion, pancreatic β-cell viability, GSK3β phosphorylation, lipolysis and glucose uptake was measured. In vivo studies were performed using KK-Ay mice, given the extract for several weeks. IPGTT was performed, and plasma insulin, FFA, food consumption and body weight were measured. In addition, diabetic KK-Ay mice were given a single dose of the extract, and IPGTT was performed.

Results

Sarcopoterium spinosum extract increased basal and glucose/forskolin-induced insulin secretion in RINm cells, and increased cell viability. The extract inhibited lipolysis in 3T3-L1 adipocytes, and induced glucose uptake in these cells as well as in AML-12 hepatocytes and L6 myotubes. GSK3β phosphorylation was also induced in L6 myotubes, suggesting increased glycogen synthesis. Sarcopoterium spinosum extract had a preventive effect on the progression of diabetes in KK-Ay mice. Catechin and epicatechin were detected in Sarcopoterium spinosum extract using hyphenated LC–MS/MS.

Conclusions

Sarcopoterium spinosum extract has effects that mimic those of insulin and provide the basis for antidiabetic activity of the extract.  相似文献

Ethnopharmacological relevance

Sarcopoterium spinosum is an abundant plant in Israel, used by Bedouin medicinal practitioners for the treatment of diabetes. In our previous study we validated the anti-diabetic activity of Sarcopoterium spinosum. The aim of this study was to further clarify its mechanism of action.

Materials and methods

In-vivo studies were performed on KK-a/y mice given the extract for 6 weeks. Insulin tolerance test was performed, and relative pancreatic islets area was measured. Mechanisms of action were investigated in L6 myotubes using protein array, Western blot analysis and confocal microscopy. Glucose uptake assays were performed in 3T3-L1 adipocytes.

Results

Sarcopoterium spinosum extract reduced fasting blood glucose and improved insulin sensitivity in treated mice. Hypertrophic islets were detected in diabetic, but not in Sarcopoterium spinosum-treated mice. Sarcopoterium spinosum phosphorylated PTEN on ser380 and thr382/383, which are known inhibitory sites. PKB was not phosphorylated by Sarcopoterium spinosum, however, translocation of PKB from cytoplasm to the membrane and nucleus was detected. Target proteins of PKB were regulated by Sarcopoterium spinosum; GSK3β was phosphorylated and cytosolic localization of FoxO was increased. Glucose uptake was increased in a PI3K and AMPK-independent mechanism.

Conclusions

We suggest that Sarcopoterium spinosum inhibited PTEN and activated PKB by a mechanism which is independent of ser473 and thr308 phosphorylation. Other post translation modifications might be involved and should be analyzed further in order to understand this unique PKB activation. Identifying the active molecules in the extract, may lead to the development of new agents for the treatment of insulin resistance.  相似文献

Aim of study

Extracts of Pistacia integerrima galls have been dispensed by Traditional Practitioners of Subcontinent for chest diseases as well as for aches and pains in the body. This study was planned to evaluate the possible analgesic and antiinflammatory effects of Pistacia integerrima extracts.

Materials and methods

Analgesia was determined using acetic acid induced abdominal constriction and formalin induced paw licking in mice. Antinociceptive effect was observed by thermally induced algesia in mice.

Results

Pistacia integerrima leaves extracts showed significant response against chemically induced pain (P < 0.001) whereas galls extracts had highly significant protection (P < 0.0001) in a dose dependent manner. In thermally induced algesia, Pistacia integerrima galls extracts 200 mg/kg (p.o.), showed significant (P < 0.05) response but less than pentazocine and diclofenac, positive references. The extracts of Pistacia integerrima 50–200 mg/kg (p.o.) had modest activity against hind paw acute and chronic inflammation induced by formalin (P < 0.01).

Conclusion

These results demonstrate that Pistacia integerrima extracts have antinociceptive and analgesic effects and no apparent acute toxicity on oral administration.  相似文献

Aim of the study

Determination of pharmacological activity relevant to wound healing of Bridelia ferruginea leaf, a traditional medicine used to treat wounds in rural Nigeria.

Materials and methods

Aqueous and ethanolic leaf extracts were tested against bacterial species of relevance to wound infections: Staphylococcus aureus, Bacillus subtilis, Escherichia coli and Pseudomonas aeruginosa. The ethanolic extracts were assessed for their ability to stimulate the growth of human dermal fibroblasts (FS5) and protect against damage induced by hydrogen peroxide. Antioxidant activity was also assessed using the DPPH assay.

Results

Both aqueous and ethanolic extracts had weak antibacterial activity (MIC > 470 μg/ml). A significant effect (p < 0.001) on the growth of FS5 fibroblasts was observed only at a concentration of 5 μg/ml (28% increase), above which the extracts appeared toxic to the cells. The ethanolic extract offered the highest protection against H2O2 damage to FS5 cells, comparable with catalase (82% at 250 μg/ml). The DPPH assay revealed antioxidant activity of the ethanolic leaf extract with IC50 12.5 ± 0.3 μg/ml comparable to l-ascorbic acid (7.3 ± 0.1 μg/ml).

Conclusion

The antibacterial, modest fibroblast stimulation activity and relatively strong antioxidant activity lend some support to the topical use of Bridelia ferruginea leaf for wound-healing in the traditional medicine of South-western Nigeria.  相似文献

Ethnopharmacological relevance

Several Searsia species (Anacardiaceae), including Searsia dentata and Searsia pyroides, are used in South Africa traditional medicine to treat epilepsy. Ethanol leaf extracts of these plants have been shown to act as possible antagonists of N-methyl-d-aspartate (NMDA)-type glutamate receptors.

Materials and methods

Leaf material of three Searsia species were collected from the Botanical Garden at the University of KwaZulu-Natal, Pietermaritzburg; dried and extracted with ethanol in an ultrasound bath. Filtered and dried extracts were resuspended in DMSO (100 mg/ml) and diluted in the recording solution. The effect of Searsia dentata, Searsia pyroides and Searsia glauca extracts was investigated in dissociated cerebellar granule cells (CGCs) from 8-day-old rats and in transiently transfected HEK (human embryonic kidney) 293 cells (HEK), expressing either NR1a/NR2A or NR1a/NR2B receptors. In both systems we measured whole-cell currents elicited by 0.5 mM NMDA (CGCs) or 50 μM glutamic acid (HEK) at −60 mV in 0 Mg and 30 μM glycine and NMDA driven Ca influx in Fura2-loaded CGC.

Results

Searsia dentata and Searsia pyroides ethanol extracts caused a dose-dependent decrease of NR current with ED50 close to 0.03 mg/ml in CGC and a similar inhibition (80% with 1 mg/ml) in HEK cells, while Searsia glauca was much less effective. The inhibition was dependent on time of incubation and slightly favored by opening of the NR channel. It was hardly reversible during the recording time, but was not caused by accelerated run-down or by interaction with the modulatory redox site. Searsia pyroides ethanol extract also depressed the NMDA stimulated increase in intracellular Ca.

Conclusions

The data confirm the specificity of Searsia dentata and Searsia pyroides and justify their use in traditional medicine. These plants may combine one or more γ-aminobutyric acid (GABA)A agonists with one or more NMDA antagonists, thus representing an efficient treatment for epilepsy.  相似文献

Aim of the study

The present study was aimed to investigate the anti-diabetic potential of the leaves of Tetracera scandens Linn. Merr. (Dilleniaceae) in vivo with regard to prove its efficacy by local herbalists in the treatment of diabetes frailties.

Materials and methods

Crude aqueous (AQ) and methanol (MEOH) extracts of the leaves of T. scandens L. were administered to both normal and alloxan induced diabetic male albino rats (Wistar strain). The blood glucose levels were measured at 0, 2, 4, 6 and 8 h after oral administration of AQ and MEOH extracts.

Results

Significant reduction in glucose was observed in fasting blood glucose levels in the treated diabetic rats without causing any hypoglycemic effect compared to normal rats. Both polar extracts of the leaves of T. scandens L. exhibited significant anti-hyperglycemic activity at different doses and intervals. The highest anti-hyperglycemic effect (62.5%) was observed by the AQ extract at 0.25 g/kg body weight (b.w.) and MEOH extract (36.5%) at 0.5 g/kg b.w. after 8 h. The significant anti-hyperglycemic activity was found to be comparable with a known oral synthetic hypoglycemic drug, glibenclamide 0.25 mg/kg b.w.

Conclusion

This study provides scientific evidence that the leaves of T. scandens L. have anti-diabetic efficacy and justifies its utility by local herbalists. However, more experiments at the clinical levels are required to confirm the utility of this plant by traditional practitioners in the management of diabetes mellitus.  相似文献

The present study was designed to investigate the hypoglycemic and hypolipidemic properties of Passiflora incarnata Linn. leaves which are widely used as traditional treatment for diabetes mellitus.

Materials and methods

The methanolic extracts of leaves of Passiflora incarnata were administered orally (100 and 200 mg/kg, for 15 days) to streptozotocin-induced diabetic mice. Hypoglycemic effects, oral glucose tolerance test, change in body weight and lipid profile of diabetic mice treated with methanolic extracts were assessed and compared with normal, diabetic control and standard drug treated mice. Histological examination during 15 days of treatment was also carried out.

Results

Methanolic extract (200 mg/kg) produced a significant reduction in fasting blood glucose level in streptozotocin-induced diabetic mice. Significant differences were also observed in urine glucose level, oral glucose tolerance test, serum lipid profile and body weight of methanolic extract treated diabetic mice, when compared with diabetic, normal and standard drug treated mice. Histopathological studies of the pancreas showed comparable regeneration of the cells by extract which were earlier necrosed by streptozotocin.

Conclusion

Methanolic extract of Passiflora incarnata exhibit significant anti-hyperglycemic and hypolipidemic activities in streptozotocin-induced diabetes in mice.  相似文献

Aim of the study

This experiment studied two extracts of Opuntia streptacantha, a plant used by the Mexican population to treat type 2 diabetes, in different assays to contribute to the understanding of the hypoglycemic mechanism of this plant.

Materials and methods

Two different extracts were prepared and tested: the first extract was a filtrate of the traditional liquefied extract (LE) preparation of the cladode; and the second filtrate extract (FE) is a filtered sample of the first. Both extracts contained a newly identified compound for Opuntia (4-hydroxy)-phenyl acetic acid derivate, they were tested on streptozotocin (STZ)-diabetic rats in a series of two tests. The first test was performed to confirm if STZ-diabetic rats presented a hypoglycemic effect after administration of the extracts (LE 135 mg/kg and FE 27 mg/kg). In the second experiment, the extracts were administered before an oral glucose tolerance test (OGTT) to confirm if they have an anti-hyperglycemic effect (LE 135 mg/kg, FE 12 and 27 mg/kg).

Results

The extracts administered to STZ-diabetic rats did not produce a significant hypoglycemic effect compared to the control group, while the same extracts administered before an OGTT produced an anti-hyperglycemic effect compared to the control group.

Conclusions

The filtered, traditional LE of the cladode of Opuntia streptacantha produces an anti-hyperglycemic effect when administered before a glucose challenge, and this anti-hyperglycemic effect is maintained after filtering the extract.Administration of both plants can improve glycemic control by blocking the hepatic glucose output, especially in the fasting state. These data support the traditional use of the plants as “agua de uso”, a cold infusion of the plant consumed over the course of a day.  相似文献

Aim of the study

Hydro-alcoholic extracts of Centaurium erythraea Rafn (CE), Gentianaceae and Artemisia herba-alba Asso (AHA), Asteraceae, medicinal plants used in traditional treatment of diabetes in north-eastern Algeria, were tested in established type 2 diabetes induced with a standardized high fat diet (HFD) in mice.

Materials and methods

After confirmation of diabetes (17th week), plant extracts were administered orally by gavage at a dose of 2 g/kg daily for 18 weeks to male C57BL/6J mice fed HFD. Animals were weighed, food intake and plasma glucose measured weekly, insulin and lipid profile at study end.

Results

At 35 weeks, groups treated with AHA or CE vs. HFD control had a significant reduction in mean (±SD) fasting blood glucose concentrations (143.8 ± 23.9 and 139.5 ± 14.2 vs. 229.0 ± 20.8 mg/dL, p < 0.05, respectively), triglyceride (18.9 ± 11.1 and 16.0 ± 6.5 vs. 62.8 ± 18.3 mg/dL, p < 0.05), total cholesterol (1.2 ± 0.1 and 1.2 ± 0.3 vs. 1.8 ± 1.1 g/L, p < 0.05) and serum insulin concentrations (1.7 ± 0.7 and 0.9 ± 0.7 vs. 3.3 ± 14.3 ng/mL, p < 0.05). Plant extracts also markedly reduced insulin resistance as compared to HFD controls (AHA: 15.6 ± 9.1, CE: 9.0 ± 7.7 vs. HFD control 38.5 ± 30.3, p < 0.05). The plant extracts decreased calorie intake and had little effect on body weight or HDL-cholesterol.

Conclusion

AHA has already been shown to have a antihyperglycaemic and antihyperlipidemic effect but this is the first demonstration of an effect of AHA and CE on established HFD-induced diabetes.  相似文献

Ethnopharmacological relevance

Rosmarinic acid (RA), a caffeic acid-related compound found in high concentrations in Prunella vulgaris (self-heal), and ursolic acid (UA), a pentacyclic triterpene acid concentrated in Salvia officinalis (sage), have been traditionally used to treat inflammation in the mouth, and may also be beneficial for gastrointestinal health in general.

Aim of the study

To investigate the permeabilities of RA and UA as pure compounds and in Prunella vulgaris and Salvia officinalis ethanol extracts across human intestinal epithelial Caco-2 cell monolayers.

Materials and methods

The permeabilities and phase II biotransformation of RA and UA as pure compounds and in herbal extracts were compared using Caco-2 cells with HPLC detection.

Results

The apparent permeability coefficient (Papp) for RA and RA in Prunella vulgaris extracts was 0.2 ± 0.05 × 10−6 cm/s, significantly increased to 0.9 ± 0.2 × 10−6 cm/s after β-glucuronidase/sulfatase treatment. Papp for UA and UA in Salvia officinalis extract was 2.7 ± 0.3 × 10−6 cm/s and 2.3 ± 0.5 × 10−6 cm/s before and after β-glucuronidase/sulfatase treatment, respectively. Neither compound was affected in permeability by the herbal extract matrix.

Conclusion

RA and UA in herbal extracts had similar uptake as that found using the pure compounds, which may simplify the prediction of compound efficacy, but the apparent lack of intestinal glucuronidation/sulfation of UA is likely to further enhance the bioavailability of that compound compared with RA.  相似文献

Ethnopharmacological relevance

Schinus molle var. areira L. (Anacardiaceae) is employed in herbal medicine for many conditions, including respiratory, urinary and menstrual disorders, and as a digestive stimulant, diuretic, astringent and antidepressant. It is also known for its topical use as wound healer, antiseptic, for skin disorders and as repellent and insecticide. In the present work, the acute dermal exposure to ethanolic and hexanic extracts from leaves of Schinus molle var. areira was studied in rats.

Materials and methods

A single dose of 2000 mg/kg of body weight of ethanolic and hexanic extracts from leaves was applied on the shaved skin of male and female rats. After 24 h of exposure, the patch was removed and any sign of irritation was recorded. Behavioral and functional parameters in a functional observational battery and motor activity in an open field were assessed after the exposure to the extracts. Then, after 14 days of observation, animals were retested. Finally, histopathological studies were conducted on several organs.

Results

Slight signs of erythema and edema were observed in the skin site of exposure, but they disappeared after 48 h. The exposure to the hexanic extract produced an increase in parameters of activity, rearing and arousal assessed in the functional observational battery, which reversed after 14 days. On the other hand, the ethanolic extract caused an increase in locomotor activity, reflected in a higher number of rearings performed in the open field in the evaluation carried out on Day 14. No histopathological alterations were detected in the analyzed organs.

Conclusions

The results show that the acute dermal exposure of the ethanolic and hexanic extracts from leaves of Schinus molle var. areira only causes a slight and reversible skin irritation, and a mild stimulatory effect in rats. All these indicate that the topical use of these extracts would be safe.  相似文献

Background

Type II diabetes and obesity are major health problems worldwide and aboriginal peoples are particularly at risk. To address this problem in Canadian native populations who find modern pharmaceuticals culturally inappropriate, our team is testing the traditional pharmacopeia of the James Bay Cree for anti-diabetic and anti-obesity activities. More specifically, the aim of the present study was to define the effects of traditional plants on intestinal glucose absorption, an under-appreciated anti-hyperglycaemic and anti-obesity activity.

Methods

Crude ethanol extracts of 17 Boreal forest medicinal plants were tested in vitro using the Caco-2 human enterocytic cell line and in vivo using an oral glucose tolerance test.

Results

Thirteen of seventeen extracts were observed to significantly inhibit uptake when administered simultaneously with 3H-deoxyglucose. Inhibition was dose-dependent and, in a few cases, even surpassed that induced by a combination of the positive controls. To validate these effects in vivo, four plant extracts were administered by intragastric gavage at 250 mg/kg to normal rats simultaneously with a 3 g/kg bolus of glucose. This resulted in a decrease in peak glycaemia by approximately 40% for two of them. Similarly, only 2 extracts reduced glucose transport after long term incubation and this could be related to reductions in the expression of SGLT-1 or GLUT-2 proteins.

Conclusions

These findings indicate that competitive inhibition of intestinal glucose uptake can be achieved by crude extracts of medicinal plants. Such extracts could be taken with meals to control postprandial glycaemia and reduce caloric intake in high risk populations that are positively inclined towards traditional medicine.  相似文献

Aim of the study

Witheringia solanacea is a small shrub that belongs to the Solanaceae family. The plant is used as an antidiabetic in Costa Rican herbal medicine. The aim of this study was to evaluate the hypoglycemic and antihyperglycemic activity of the aqueous extract of W. solanacea leaves in rodent models.

Materials and methods

A crude extract of W. solanacea leaves was prepared in boiling water and the aqueous filtrate was lyophilized. A single oral dose of 250, 500 and 1000 mg/kg of the extract was evaluated for hypoglycemic activity in a glucose tolerance test in normal rats and for antihyperglycemic activity in alloxan-induced (140 mg/kg) diabetic rats. The blood glucose level was determined at different times by the glucose oxidase method.

Results

Dosage of 500 and 1000 mg/kg of the extract significantly decreased (p < 0.05) blood glucose levels in the glucose tolerance test in normal rats after 1 h, there was no significant difference observed at 250 mg/kg. Dose of 500 mg/kg of the extract significantly reduced (p < 0.05) blood glucose levels in alloxan induced hyperglycemic rats at 4 and 5 h.

Conclusions

In the present study, the hypoglycemic and antihyperglycemic potential of the W. solanacea was demonstrated in rats. These results give support to the traditional use of W. solanacea as antidiabetic herbal medicine.  相似文献

Ethnopharmacological importance

Many species of plants in the Brazilian cerrado (savanna) are widely used in ethnomedicine. However, the safety and effectiveness of medicinal plants used in communities with little or no access to manufactured drugs should be evaluated.

Aim of the study

Evaluate the antimicrobial and cytotoxic activities of extracts from eight plant species, obtained using Brazilian cachaça as the extractor liquid.

Materials and methods

The extracts were tested against Bacillus subtilis, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans, Candida parapsilosis, promastigote forms of Leishmania amazonensis, and poliovirus. In addition, cytotoxic activity was assayed in Vero cells and in human erythrocytes.

Results

The plant species Curatella americana, Sclerolobium aureum, and Plathymenia reticulata showed the best activity against yeasts, especially the crude extract of C. americana and its ethyl-acetate fraction. Kielmeyera lathrophyton showed a minimum inhibitory concentration of 250 μg/ml against S. aureus, and was inactive against Gram-negative bacteria. The extract obtained from Annona coriacea showed the best activity against the promastigote forms of Leishmania amazonensis (IC50 = 175 μg/ml). Only C. americana showed potential for antipoliovirus activity. The concentrations of the crude extracts that showed toxicity to VERO cells had CC50 between 31 and 470 μg/ml, and the lyophilized Brazilian cachaça showed a CC50 of 307 μg/ml. None of the extracts showed toxicity against human erythrocytes.

Conclusions

Among the plant species studied, C. americana proved to be effective against microorganisms, especially as an antifungal. The results will help in the search for alternative drugs to be used in pharmacotherapy, and will contribute to establish safe and effective use of phytomedicines in the treatment of infectious diseases.  相似文献

Ethnopharmacological relevance

Glycosmis arborea Roxb, syn. Glycosmis pentaphylla Retz, (Rutacea) is a shrub found all over India, have been used for the healing of wounds of livestock in Indian folk medicine.

Aim of the study

The present study provides a scientific evaluation for the wound healing potential of ethanolic extract of Glycosmis arborea leaves.

Methods and materials

Excision and incision wounds were inflicted upon four groups containing six rats each. Group-1 was assigned as control (ointment base), Group-2 and Group-3 were treated with 5 and 10% EtOH extract ointment. Group-4 was treated with standard Nitrofurazone (0.2%) ointment. The parameters observed were percentage of wound contraction, epithelization period, tensile strength, hydroxyproline content. Ethanolic extract of Glycosmis arborea leaves was given orally at a dose of 200, 400 mg/kg/day in dead space wound model.

Results

Topical application of Glycosmis arborea ointment (10%) in excision wound model significantly (P < 0.05) increased the percentage of wound contraction (96.91%) compared with control (60.27%) and decreased the epithelization time. The tensile strength significantly (P < 0.05) increased in Group-3 at 560.33 ± 6.48 g when compared to control at 319.17 ± 6.16 g. Rats treated with Glycosmis arborea extract (400 mg/kg) showed significant (P < 0.05) increase in hydroxyproline content at 54.94 ± 0.96 mg/g when compared with control at 30.77 ± 1.13 mg/g.

Conclusion

The ethanolic extract of Glycosmis arborea facilitated wound healing significantly, corroborating the folk medicinal use of this plant.  相似文献

Aim

Symplocos cochinchinensis (Lour.) S. Moore. is used in Indian system of traditional medicine to treat diabetes mellitus. The present study aims to investigate the antidiabetic efficacy of the hexane extract of Symplocos cochinchinensis leaves in high fat diet-low streptozotocin (STZ) induced type 2 diabetic rats.

Materials and methods

The doses for the study were fixed based on Irwin test. The hypoglycemic effect of the hexane extract of Symplocos cochinchinensis leaves were studied in normal rats. Oral glucose and insulin tolerance tests were carried out. The antihyperglycemic effect of the hexane extract at 250 and 500 mg/kg was studied in high fat diet-low STZ induced type 2 diabetic rats for 28 days.

Results

The extracts showed no adverse effects up to 5 g/kg concentration. In hypoglycemic study, after treatment with hexane extract at 250 and 500 mg/kg the blood glucose was mildly reduced. In oral glucose tolerance test, the treatment with the hexane extract at 250 and 500 mg/kg showed a highly significant reduction of 12.07% and 23.58% in plasma glucose levels, respectively 30 min after glucose load. The insulin tolerance test also showed improved insulin sensitivity after 60 min of insulin treatment. In high fat diet-low STZ induced type 2 diabetic rats, after 28 days treatment with the hexane extract at 250 and 500 mg/kg reduced the plasma glucose level by 17.04% and 42.10%, respectively. A significant reduction in plasma insulin, plasma and hepatic total cholesterol (TC), triglycerides (TG) and free fatty acids (FFA) and a significant increase in liver glycogen were observed in treated diabetic rats.

Conclusion

This study demonstrated the potential antidiabetic property of hexane extract of Symplocos cochinchinensis leaves on type 2 diabetes mellitus, thus justifying its traditional usage.  相似文献

For identification of the active constituents we investigated the anticancer activity of cardenolides from Streptocaulon tomentosum Wight & Arn. (Asclepiadaceae) and from Nerium oleander L. (Apocynaceae) which are both used against cancer in the traditional medicine in their region of origin.

Material, methods and results

The antiproliferative activity of cardenolides isolated from roots of Streptocaulon tomentosum (IC50 < 1-15.3 μM after 2 days in MCF7) and of cardenolide containing fractions from the cold aqueous extract of Nerium oleander leaves (“Breastin”, mean IC50 0.85 μg/ml in a panel of 36 human tumor cell lines), their influence on the cellular viability and on the cell cycle (block at the G2/M-phase or at the S-phase in tumor cells, respectively) were determined using different cell lines. The murine cell line L929 and normal non-tumor cells were not affected. Bioactivity guided fractionation of Breastin resulted in the isolation of the monoglycosidic cardenolides oleandrine, oleandrigeninsarmentoside, neritaloside, odoroside H, and odoroside A (IC50-values between 0.010 and 0.071 μg/ml).

Conclusions

The observed anticancer activities of extracts and isolated cardenolides are in agreement with the ethnomedicinal use of Streptocaulon tomentosum and Nerium oleander. The most active anticancer compounds from both species are monoglycosidic cardenolides possessing the 3β,14β-dihydroxy-5β-card-20(22)-enolide structure with or without an acetoxy group at C-16. The results indicate that the cytotoxic effects are induced by the inhibition of the plasma membrane bound Na+/K+-ATPase.  相似文献

Ethopharmacological relevance

Coreopsis tinctoria flowering tops infusion is traditionally used in Portugal for treating the symptoms of diabetes. Recent studies have revealed its antihyperglycemic activity when administered for 3 weeks to a STZ-induced glucose intolerance model in the rat and glucose tolerance regain was even clearer and pancreatic function recovery was achieved when administering Coreopsis tinctoria flavonoid-rich AcOEt fraction.In this study we aimed to evaluate the protective effect of Coreopsis tinctoria flowering tops aqueous extract, AcOEt fraction and the pure compounds marein and flavanomarein, against beta-cell injury, in a mouse insulinoma cell line (MIN6) challenged with pro-oxidant tert-Butyl-Hydroperoxide (tBHP) or cytokines.

Materials and methods

The protective effects of Coreopsis tinctoria flowering tops extracts and pure compounds were evaluated through pre-incubating MIN6 cells with samples followed by treatment with tBHP (400 μM for 2 h) after which viability was determined through ATP measurements. In order to assess whether plant extracts were involved in decreasing reactive oxygen species, superoxide anion production was determined through a lucigenin-enhanced chemiluminescent method. Lastly, the direct influence of Coreopsis tinctoria extracts and main compounds on cell survival/apoptosis was determined measuring caspase 3 and 7 cleavage induced by cytokines.

Results

Coreopsis tinctoria flowering tops extracts (25-100 μg/mL) and pure compounds (200-400 μM), when pre-incubated with MIN6 cells did not present any cytotoxicity, instead they increased cell viability in a dose dependent manner when challenged with tBHP. Treatment with this pro-oxidant also showed a rise in superoxide radical anion formation in MIN6 cells. This increase was significantly reduced by treatment with superoxide dismutase enzyme (SOD) but not by pre-treatment with Coreopsis tinctoria flowering tops extracts. Caspase 3/7 activation measurements show that Coreopsis tinctoria flowering tops extracts, as well as marein and flavanomarein, significantly inhibit apoptosis.

Conclusions

Coreopsis tinctoria extracts and pure compounds show cytoprotection that seems to be due to inhibition of the apoptotic pathway, and not through a decrease on superoxide radical production.  相似文献

Ethnopharmacological relevance

In the folk-traditional medicine, snails were used to purify blood, boost immune system, prevent conjunctivitis and to treat liver problems.

Objectives

To evaluate the hepatoprotective activity of the edible snail (Bellamia bengalensis) flesh extract in male Wistar rats treated with carbon tetrachloride as an hepatotoxicant.

Materials and methods

Live adult Bellamia bengalensis was collected commercially from the Kolkata market. Aqueous flesh extract (BBE) was prepared in 0.9% saline and expressed in terms of wet weight basis. The aqueous flesh extract was administered orally (1, 2 g kg−1 day−1) to male rats for 12 days. Liv52 was used as positive control. 24 h after administration of extract, the rats were given a single oral dose of CCl4 (1.25 ml kg−1), except vehicle control rats. After 24 h of CCl4 administration, all the animals were sacrificed to collect the blood and liver tissue.

Results

BBE (1 and 2 g kg−1 day−1, p.o. × 12 days) significantly prevented CCl4 induced elevation of SGOT, SGPT, γGT, ACP, ALP, bilirubin, LDH and CCl4 induced decrease in total protein, triglyceride level in male Wistar rats. BBE treated rat liver anti-oxidant parameters (LPO, SOD, GSH, CAT, GPx) were significantly antagonized for the pro-oxidant effect of CCl4. Histopathological studies also supported the protective effect of BBE.

Conclusion

This study validated the folk and traditional use of snail in liver disorder through CCl4-induced rat experimental model.  相似文献

Ethnopharmacological relevance

Five medicinal plants including Anogeissus acuminata (Roxb. ex DC.) Gills. & Perr. (Combretaceae), Catunaregam tormentosa (Bl. ex DC.) Tirveng (Rubiaceae), Dioecrescis erythroclada (Kurz) Tirveng. (Rubiaceae), Mimosa pudica Linn. var. hispida Bren. (Fabaceae), and Rauwolfia serpentina (L). Benth. ex Kurz. (Apocyanaceae), which have been traditionally used for the treatment of diabetes mellitus and other diseases for several generations by the Thai-Lanna people in the Northern part of Thailand were investigated for their hypoglycemic activity in normoglycemic and alloxan, induced diabetic mice.

Materials and methods

The aqueous extracts of the selected five medicinal plants were tested for their phytochemicals, free radical scavenging activity and hypoglycemic activity on 18 h fasted normoglycemic and alloxan induced diabetic mice over a period of 4 h comparing with the standard anti-diabetic drugs (insulin and glibenclamide) using two way analysis of variance (ANOVA) as analytical tool. Phytochemical analysis was performed using the standard methods while 2,2-diphenyl-1-picrylhydrazine (DPPH) was used to test for free radical scavenging activities of the medicinal plant extracts.

Results

Phytochemicals detected in the extracts were glycosides, xanthones, tannins, alkaloids and saponins. Anogeissus acuminata showed the highest free radical scavenging activity with the IC50 value of 11.00 μg/mL which was 4 folds of the standard ascorbic acid. Significant reduction in fasting blood glucose (FBG) levels of the normoglycemic mice was observed at 4 and 3 h with the extracts of Mimosa pudica (200 mg/kg bw) and Rauwolfia serpentina (100 mg/kg bw), and percentage decreases of 46.15 and 27.94% which were 0.76 and 1.47; 0.53 and 0.91 folds of insulin and glibenclamide, respectively. In alloxan induced diabetic mice, all extracts showed significant (p < 0.05) hypoglycemic activity, with the maximum FBG reduction of 78.96 at 100 mg/kg bw shown by Anogeissus acuminata at the 4 h. The hypoglycemic activity of Anogeissus acuminata was comparable to insulin (1.1 fold), but more potent than glibenclamide (1.76 folds).

Conclusions

Medicinal plants selected from the Thai/Lanna Medicinal Plant Recipe Database MANOSROI II showed hypoglycemic activity in normoglycemic and alloxan induced diabetic mice. This study confirmed the traditional use of these medicinal plants for the treatment of diabetes mellitus and the thiazolidiendiones mimic hypoglycemic effects of the medicinal plants was suggested.  相似文献
