ObjectiveMatrix metalloproteinases (MMPs), particularly MMP-2 and MMP-9, have been reported as putative tumor markers because of their involvement in cancer invasion and metastasis. The aim of our study was to elucidate the possible role of MMP-2 and -9 as serum prognostic biomarker for breast cancer classification and correlate it with the clinicopathological variables.Design and MethodsOur study consisted of 60 females with primary breast cancer, 40 cases of benign breast disease and 60 healthy female volunteers as controls. The serum MMP-2 and -9 levels were quantitatively measured by ELISA technique.ResultsA significantly raised MMP-2 and MMP-9 levels were observed in breast cancer patients. Significant rise in serum MMP-9 concentration was found in patients presenting with metastasis as well as in those cases who presented with a duration of less than 1 year. ROC analyses depicted a serum cutoff value of 315 ng/mL for MMP-9 to discriminate the breast cancer patients from the control group.ConclusionOur results suggest that serum MMP-9 level is a better marker than serum MMP-2 in predicting the breast cancer development and progression.  相似文献

目的:检测基质金属蛋白酶-3(MMP-3)和细胞外基质金属蛋白酶诱导因子(EMMPRIN)在乳腺癌演变过程中的表达并探讨其意义。方法:应用免疫组化二步法,检测58例乳腺癌、28例非典型导管增生、40例普通导管增生及20例正常乳腺组织中MMP-3和EMMPRIN蛋白的表达,并采用原位杂交法检测各种病变及正常乳腺组织各20例中MMP-3mRNA、EMMPRIN mRNA的表达。结果:MMP-3、EMMPRIN及MMP-3mRNA、EMMPRIN mRNA在乳腺癌组织(16.98±1.47,20.42±1.54,12.88±1.34,12.30±1.25)和非典型导管增生组织(19.05±1.38,17.38±1.69,13.18±1.34,13.49±1.25)中的IOD值较普通导管增生组织(10.96±1.20,10.23±1.09,10.47±1.12,10.23±1.09)及正常乳腺组织(10.36±1.05,10.22±1.20,10.47±1.14,10.21±1.06)高(P<0.01)。MMP-3、EMMPRIN和MMP-3 mRNA、EMMPRIN mRNA在乳腺癌和非典型导管增生组织中的表达水平...  相似文献

目的 探讨肠复康对人结肠癌 HT29裸小鼠移植瘤基质金属蛋白酶- 2 (MMP-2)、金属蛋白酶组织抑制因子- 2( TIMP-2)的影响 ,以此对肿瘤康复领域的功能性改良措施的选择提供有利依据. 方法 建立人结肠癌 HT29癌细胞株裸小鼠皮下移植瘤模型,采用随机数字表法随机分成 3组,模型组 8只,肠复康组 7只,西药组 8只.免疫组化染色结合图像分析系统半定量检测移植瘤组织内 MMP-2、 TIMP-2积分光密度( IOD)及 MMP-2/TIMP-2比值. 结果 与模型组比,肠复康组可使移植瘤 MMP-2和 TIMP-2含量增高,前者无显著差异 (P >0.05),后者具明显差异 (P< 0.01),西药组也能增高移植瘤 MMP-2和 TIMP-2含量,但皆无明显差异 (P >0.05);肠复康组和西药组可降低 MMP-2/TIMP-2比值,前者有显著差异 (P< 0.05),后者无明显差异 (P >0.05). 结论 肠复康可降低移植瘤内 MMP-2/TIMP-2比值,对人结肠癌 HT29裸小鼠移植瘤细胞外基质( ECM)降解可能具有抑制作用.  相似文献

目的:通过检测不稳定性心绞痛患者外周血基质金属蛋白酶-1、白介素-1β、C反应蛋白水平的变化,探讨基质金属蛋白酶-1在不稳定性心绞痛患者危险度分层以及预后价值中的作用。方法:选择不稳定性心绞痛患者(UA组)32例,稳定性心绞痛患者(SA组)28例及健康对照组20例,采用酶联免疫吸附法(ELISA)分别测定各组外周血基质金属蛋白酶-1及白介素-1β的水平,采用免疫比浊法测定各组C反应蛋白的水平,分析UA组基质金属蛋白酶-1水平与心脏事件发生率及C-反应蛋白之间的相关性。结果:UA组基质金属蛋白酶-1水平、白介素-1β、C-反应蛋白水平显著高于SA组(P〈0.01)及对照组(P〈0.01);UA组基质金属蛋白酶-1水平与C-反应蛋白之间呈正的直线相关(r=0.489,P〈0.01),UA组基质金属蛋白酶-1水平升高者发生心脏事件的几率明显高于水平低者(P〈0.05)。结论:基质金属蛋白酶-1可能是不稳定性心绞痛患者危险度分层及预后的有价值的生化指标。  相似文献

In the present study, we investigated the antitumor effects of the invasiveness and migration of heme oxygenase 1 (HO-1) in human breast carcinoma cells. 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced matrix metalloproteinase-9 (MMP-9) enzyme activity and gene expression at both protein and mRNA levels were examined in human breast carcinoma cells (MCF-7 and MDA-MB-231), and the addition of the MMP-9 inhibitor, SB3CT, significantly suppressed TPA-induced invasion and migration according to the in vitro Transwell assay. Elevation of HO-1 gene expression by ferric protoporphyrin IX inhibited TPA-induced invasion of MCF-7 cells, which was blocked by adding the heme oxygenase inhibitor, tin protoporphyrin IX, or transfection of cells with HO-1 short hairpin RNA. MCF-7 cells overexpressing HO-1 (MCF-7/HO-1) were established in the present study, and TPA-induced MMP-9 gene expression, tumor invasion, and colony formation were significantly reduced in MCF-7/HO-1 cells, compared with those in Neo-transfected cells. Activation of protein kinase Calpha/extracellular signal-regulated kinases/AP-1 with stimulation of reactive oxygen species production was involved in TPA-induced invasion of MCF-7 cells, which was attenuated by HO-1 protein induced by ferric protoporphyrin IX or transfection of HO-1 expression vectors. Additionally, the addition of carbon monoxide, but not ferric ions, biliverdin, or bilirubin, inhibited TPA-induced invasion through suppressing MMP-9, extracellular signal-regulated kinases, and AP-1 activation stimulated by TPA. The beneficial role of HO-1 in blocking tumor invasion was first identified in this study.  相似文献

目的探讨乳腺癌及其术后和乳腺良性肿瘤患者与血清同型半胱氨酸(Hcy)关系。方法采用循环酶法检测90例乳腺癌、乳腺癌术后及乳腺良性肿瘤患者血清Hcy水平,并与50例健康体检者(健康对照组)检测结果比较。结果乳腺癌组与乳腺良性肿瘤组、健康对照组之间血清Hcy水平的差异无统计学意义(P>0.05)。乳腺癌术后组与健康对照组之间血清Hcy水平差异有统计学意义(P<0.05)。结论血清Hcy与乳腺肿瘤的发生、发展无相关性。  相似文献

目的 研究细胞核因子(NF)-κBp50及基质金属蛋白酶(MMP-9)在乳腺癌发生、发展、转移扩散中的作用及其与乳腺癌临床病理特征的关系.方法 收集我院152例乳腺组织标本(包括乳腺癌、乳腺增生、癌旁乳腺组织),应用免疫组织化学Elivision法检测乳腺癌92例(乳腺癌组)、乳腺增生组织30例(乳腺增生组)、癌旁乳腺组织30例(癌旁组)中NF-κBp50及MMP-9蛋白的表达情况.结果 NF-κBp50、MMP-9蛋白在乳腺癌组明显高于癌旁组和乳腺增生组,其上调程度随着乳腺癌分化程度的降低、TNM分期的增加、淋巴结转移的出现及瘤块增大而逐渐增高.低分化组表达较高、中分化组显著升高.二者的表达随肿瘤体积的增大而增大,在直径≤2 cm乳腺癌中的表达明显低于直径>2 cm乳腺癌中的表达.结论 NF-κB、MMP-9在乳腺癌组织中表达上调,二者相关性好,与乳腺癌的分化程度、TNM分期、淋巴结转移的出现及瘤块的大小有关.NF-κB、MMP-9可能通过相互调控参与乳腺癌的侵袭转移.  相似文献

目的:研究口腔疣状癌中基质金属蛋白酶(MMP)-2、MMP-9及金属蛋白酶组织抑制因子(TIMP-2)的表达.探讨其在口腔疣状癌发生发展过程中的作用和意义.方法:取25例口腔疣状癌,10例正常口腔黏膜,25例口腔鳞癌(高、低分化鳞癌各为15、10例),应用免疫组织化学法检测上述标本中MMP-2、MMP-9及,TIMP-2的表达和分布.结果:正常口腔黏膜组织TIMP-2为阴性表达:MMP-2、MMP-9的阳性表达率分别为10.00%、20.00%.口腔疣状癌MMP-2阳性表达率为28.00%.MMP-9阳性表达率为44.00%,TIMP-2阳性表达率为72.00%.口腔疣状癌、口腔高分化鳞癌、口腔低分化鳞癌MMP-2、MMP-9、TIMP-2表达均高于正常口腔黏膜(P<0.05).结论:在口腔疣状癌,口腔高分化鳞癌、口腔低分化鳞癌中.MMP-2与MMP-9的阳性表达率呈正相关,与TIMP-2呈负相关;相关分析发现,MMP-2、MMP-9和TIMP-2的袁达两两相关,同时3者的表达可能单独或共同参与.  相似文献

目的探讨乳腺癌超声造影特征与血管生成拟态(VM)的相关性及在鉴别诊断中的价值。方法选取经手术病理证实为乳腺浸润性导管癌患者152例,术前均行常规超声和超声造影检查,术后标本行病理检查。记录患者临床病理特征(年龄、肿瘤大小、临床分期、是否淋巴结转移)和超声造影特征(均匀性、是否存在灌注缺损、增强强度、增强形态、边界等),分析VM阳性与阴性的乳腺癌超声造影特征的差异。绘制受试者工作特征(ROC)曲线分析各因素鉴别VM阳性的诊断效能。结果152乳腺癌患者中,经病理确诊VM阳性率为26.3%。病灶直径>2 cm、临床分期为Ⅲ期及Ⅳ期、腋窝淋巴结转移与VM阳性相关(均P<0.05)。超声造影结果显示病灶增强形态不规整、边界不清晰、增强模式为快进快退和快进慢退与VM阳性相关(均P<0.05)。其中病灶增强边界不清晰鉴别VM阳性的价值最高,曲线下面积为0.764,准确率、敏感性、特异性分别为85.0%、66.7%、90.3%。结论超声造影特征与VM阳性存在一定的相关性,超声造影表现为形态不规整、边界不清晰、快进快退征象可作为诊断乳腺癌VM阳性的有效辅助手段。  相似文献

Multiple steps are involved in the metastasis of cancer cells from primary sites to distant organs. These steps should be considered in the design of pharmacologic approaches to prevent or inhibit the metastatic process. In the present study, we have compared the effects of inhibiting several steps involved in the bone metastatic process individually with inhibition of both together. The steps we chose were matrix metalloproteinase (MMP) secretion, likely involved in tumor cell invasion, and osteoclastic bone resorption, the final step in the process. We used an experimental model in which inoculation of human estrogen-independent breast cancer MDA-231 cells into the left cardiac ventricle of female nude mice causes osteolytic lesions in bone. To inhibit cancer invasiveness, the tissue inhibitor of the MMP-2 (TIMP-2), which is a natural inhibitor of MMPs, was overexpressed in MDA-231 cells. To inhibit bone resorption, a potent bisphosphonate, ibandronate (4 microg/mouse) was daily administered subcutaneously. Nude mice received either; (a) nontransfected MDA-231 cells; (b) nontransfected MDA231 cells and ibandronate; (c) TIMP-2-transfected MDA-231 cells; or (d) TIMP-2-transfected MDA-231 cells and ibandronate. In mice from group a, radiographs revealed multiple osteolytic lesions. However, in mice from group b or group c, osteolytic lesions were markedly decreased. Of particular note, in animals from group d receiving both ibandronate and TIMP-2-transfected MDA-231 cells, there were no radiologically detectable osteolytic lesions. Survival rate was increased in mice of groups c and d. There was no difference in local enlargement in the mammary fat pad between nontransfected and TIMP-2-transfected MDA-231 cells. These results suggest that inhibition of both MMPs and osteoclastic bone resorption are more efficacious treatment for prevention of osteolytic lesions than either alone, and suggest that when therapies are designed based on the uniqueness of the bone microenvironment and combined with several common steps in the metastatic process, osteolytic bone metastases can be more efficiently and selectively inhibited.  相似文献

目的 探讨血清基质金属蛋白酶-9(MMP-9)与2型糖尿病(T2DM)患者颈动脉粥样硬化的关系.方法 选择93例T2DM患者作为研究对象,测量患者颈动脉内中膜厚度(IMT)、斑块情况.根据IMT值将选取T2DM患者分为3组:单纯糖尿病组32例(糖尿病组),糖尿病颈动脉内中膜增厚组31例(中膜增厚组),糖尿病颈动脉粥样硬化斑块形成组30例(斑块组);另选健康查体者30名作为对照组.测定血清MMP-9浓度并进行相关分析.结果 糖尿病组血清MMP-9为(550.26±269.28)μg/L,显著高于对照组(359.70±215.62)μg/L,差异有统计学意义(t=2.23,P<0.05);中膜增厚组MMP-9为(712.15±340.47)μg/L明显高于对照组(t=4.53,P<0.01)和单纯糖尿病组(t=2.40,P<0.05);斑块组MMP-9为(889.08±247.80)μg/L明显高于对照组(t=7.01,P<0.01)、糖尿病组(t=4.89,P<0.01)和中膜增厚组(t=2.53,P<0.05).结论 血清MMP-9升高与颈动脉粥样硬化的病变程度密切相关.
Abstract:
Objective To investigated the relationship between serum matrix metalloproteinase-9(MMP-9) and carotid atherosclerosis(AS) in type 2 diabetes mellitus(T2DM).Methods A total of 93 patients with T2DM were recruited to our study.The intima-media thickness(IMT) and plaques of carotid artery were measured.These patients were divided into 3 groups according to their IMT values: diabetes mellitus(DM) group(n=32),carotid artery intima thicken group(n=31) and carotid artery intima plaque group(n=30).At the same time,30 healthy individuals were selected as control.Serum level of MMP-9 were determined and analyzed.Results The serum MMP-9 in DM group was significantly higher than that in healthy controls([550.26±269.28]μg/L vs.[359.70±215.62]μg/L,t=2.23,P<0.05).The serum MMP-9 level of intima thicken group(712.15±340.47)μg/L was significantly higher than that in healthy controls(t=4.53,P<0.01) and DM group(t=2.40,P<0.05).The serum MMP-9 level of plaque group([889.08±247.80]μg/L) was even more significantly higher than DM group(t=4.89,P<0.01),IMT group(t=2.53,P<0.05) and healthy controls(t=7.01,P<0.01).Conclusion The severity of carotid atherosclerosis in T2DM is closely associated with the serum MMP-9 level.  相似文献

目的通过超声检查颈动脉硬化斑块,结合检测患者血浆高敏C-反应蛋白(hs-CRP)、基质金属蛋白酶(MMP,包括MMP-2、MMP-9)、CD40配体(CD40L)水平综合评价斑块的易损性。方法对68例首次发病的脑梗死患者(A组)、62例无症状颈动脉硬化患者(B组)和56例健康对照者(C组)进行研究,超声检查颈动脉内膜-中层厚度(IMT)、斑块类型,采用酶联免疫吸附法检测血浆hs-CRP、MMP-2、MMP-9、CD40L。A组患者进行神经功能缺损程度评分。结果 A组IMT高于B组(P=0.024),A组以易损斑块(60.3%)为主,B组以稳定斑块(77.4%)为主;A组患者血浆hs-CRP、MMP-2、MMP-9、CD40L水平均高于B组和C组(均P<0.05);A组易损斑块者血浆MMP-2、MMP-9、CD40L水平均高于稳定斑块和无斑块者(均P<0.05);易损斑块者hs-CRP与MMP-2、MMP-9均呈正相关(r=0.516,P=0.001;r=0.653,P=0.001);MMP-2与MMP-9、CD40L均呈正相关(r=0.637,P=0.001;r=0.614,P=0.010);MMP-9与CD40L呈正相关(r=0.749,P=0.012);易损斑块者神经功能缺损程度评分(26.19±11.25)高于稳定斑块(23.58±10.46)及无斑块者(23.14±10.18);血浆hs-CRP、MMP-2、MMP-9、CD40L与神经功能缺损程度评分均呈正相关(r=0.652,P=0.001;r=0.761,P=0.001;r=0.729,P=0.001;r=0.648,P=0.010)。结论超声检查颈动脉硬化斑块类型结合检测血浆相关炎症介质水平有助于评价斑块的易损性。  相似文献

目的:探讨不稳定型心绞痛(UAP)患者血清致炎因子(IL-1、IL-6、TNF-α)、抗炎因子(IL-10)水平与基质金属蛋白酶-2(MMP-2)及其抑制剂(TIMP-2)的关系.方法:选择UAP患者135例(UAP组)、稳定型心绞痛(SAP)患者35例(SAP组)、对照组30例,应用ELISA法检测血清IL-1、IL-6、TNF-α、IL-10、MMP-2、TIMP-2水平.结果:UAP组患者血清IL-1、IL-6、TNF-α、IL-10、(IL-1 +IL-6+TNF-a)/IL-10、MMP-2、TIMP-2、MMP-2/TIMP-2水平高于对照组及SAP组(P均<0.05);Spearson相关分析显示,UAP患者血清MMP-2活性与IL-1、IL-6、IL-10、TNF-α水平正相关(P<0.05);多元逐步回归分析显示,UAP患者血清MMP-2与IL-1、(IL-1+IL-6+TNF-α)/IL-10、空腹血糖独立正相关.结论:UAP患者致炎与抗炎细胞因子平衡失调,可能通过影响MMP-2活性介导斑块向不稳定性进展,促进UAP的发生.  相似文献

Corticotropin-releasing hormone (CRH), synthesized in the hypothalamus, is also produced at several extrahypothalamic sites and in normal endometrial cells. CRH exerts antiproliferative activity on oestrogen-dependent tumour cell lines (Ishikawa cells and breast cancer cells) via the CRH receptor-1. This study investigated the potential role of CRH as a factor affecting endometrial migration and invasion in Ishikawa cells, and the possible mechanisms involved in this process. Increasing concentrations of CRH (1, 10 and 100 nM) significantly reduced the proliferation of Ishikawa cells but increased the invasiveness these cells compared with the control group. All three concentrations of CRH significantly increased matrix metalloproteinase (MMP)-2 and MMP-9 levels in Ishikawa cells. In conclusion, CRH inhibited the growth of Ishikawa cells but enhanced their invasiveness, possibly by increasing MMP-2 and MMP-9 levels. These findings suggest that CRH might induce invasion and migration by upregulating MMP-2 and MMP-9 in endometrial cancer.  相似文献

[目的]探讨乳腺癌病人希望水平与社会支持的相关性。[方法]采用一般状况调查表、Herth希望量表、社会支持量表对222例乳腺癌病人进行问卷调查,运用SPSS 17.0软件包进行统计分析。[结果]乳腺癌病人的希望水平总分为36.53分±3.27分,高等希望水平的病人占55.9%,中等希望水平的占44.1%;乳腺癌社会支持总分为44.28分±6.26分,高水平社会支持的病人占48.6%,中等水平的占51.4%;希望水平与社会支持总分及各维度之间呈正相关。[结论]乳腺癌病人的希望水平与社会支持有显著的相关性,护士应调动病人的社会支持系统,提高病人战胜疾病的信心,进一步提升其生存质量。  相似文献

BACKGROUND: In the present study, we investigated the plasma levels of M-CSF and commonly accepted tumor marker (antigen CA 15-3) in breast cancer patients in relation to the group with benign breast tumor and to the healthy controls. Additionally, we compared the plasma level of M-CSF with the tumor stage of breast cancer and defined the diagnostic criteria: sensitivity, specificity, the positive and the negative predictive values. Moreover, we defined the receiver-operating characteristics (ROC) curve for M-CSF and CA 15-3, and correlation between both parameters. METHODS: M-CSF and CA 15-3 were measured in 80 patients with breast cancer, 17 patients with benign breast tumor and in 30 healthy subjects. M-CSF was determined using enzyme-linked immunosorbent assay (ELISA). CA 15-3 was measured using a microparticle enzyme immunoassay kit (MEIA). RESULTS: There were statistically significant differences in the levels of circulating M-CSF and CA 15-3 in the breast cancer patients comparing to the group with benign breast tumor and to the control group. The levels of M-CSF and CA 15-3 were also significantly higher in patients with more advanced tumor stage. Statistically significant positive correlation was observed between the M-CSF and CA 15-3 levels. The M-CSF and CA 15-3 diagnostic specificities were 95%. The diagnostic sensitivity (59%), the positive predictive value (97%) and the negative predictive value (41%) were higher for M-CSF than for CA 15-3 (48.8%, 95% and 40.1%, respectively). The combined use of both cytokines resulted in the increase of the sensitivity to the range of 70%. We observed a higher range of the diagnostic sensitivity of M-CSF in more advanced breast tumor stage. The M-CSF area under the ROC curve was larger (0.801) than the ROC area of CA 15-3 (0.785). CONCLUSIONS: These results suggest that M-CSF is the good candidate for a breast cancer tumor marker.  相似文献

目的 探讨乳腺癌的彩色多普勒超声表现与肿瘤大小、组织病理学类型的相关性.方法 回顾性分析200例乳腺癌患者202个肿瘤的彩色多普勒超声表现,分析乳腺癌血流分布类型、血流分级与其大小、组织病理学类型的相关性.结果 202个乳腺癌病灶的血流检出率为91.58%(185/202),周边型血流17.33%(35/202),内部型血流51.49%(104/202),穿入型血流22.77%(46/202),8.42%(17/202)无血流信号.非浸润性乳腺癌、浸润性导管癌、特殊类型乳腺癌之间及浸润性导管癌Ⅰ级、Ⅱ级、Ⅲ级之间血流分布类型、Adler分级的差异均无统计学意义(P均>0.05);但不同大小乳腺癌之间血流分布类型、Adler分级差异有统计学意义,肿块越大,内部型血流、穿人型血流越多,Adler分级越高(P<0.05).结论 乳腺癌血流分布类型、血流分级与大小均呈正相关.乳腺癌彩色多普勒超声表现与其组织病理学类型无明显相关性.  相似文献
