A549, a human lung cancer cell line, spontaneously produces a tumor-derived immunosuppressive factor (TDSF) which inhibits the production and action of interleukin 1 (IL-1). After exposure of macrophages to TDSF for 5 h, the production of IL-1 by macrophages was significantly inhibited. The inhibition was much stronger if TDSF existed continuously in macrophage culture. The response of thymocytes treated with nylon wool to exogenous IL-1 was significantly suppressed in the presence of TDSF, suggesting that TDSF can inhibit the action of IL-1. The thymocytes untreated with nylon wool could proliferate after being stimulated with Con A. The proliferation was significantly suppressed by TDSF in a dose-dependent manner. These results suggest that the inhibitory action of TDSF on T cell activation is associated with IL-1, and that TDSF might exert an inhibitory action on other reactions mediated by IL-1. Furthermore, TDSF can reduce the supplementation of new T cells by inhibiting the proliferation of thymocytes.  相似文献

人肺癌A549细胞产生的免疫抑制因子(TDSF)作用于巨噬细胞(Mφ)5h,即可抑制白细胞介素-1(IL-1)的产生;如持续作用24 h、48h则抑制作用更强。与对照组相比P<0.05,<0.01.当TDSF存在时,经尼龙毛处理的胸腺细胞对外源性IL-1反应显著减弱,表明TDSF能抑制IL-1作用。ConA刺激未经尼龙毛处理的胸腺细胞可产生增殖,但TDSF使其增殖抑制.表明:TDSF能抑制IL-1的产生及作用,并能干扰由IL-1介导的其他反应.TDSF可通过抑制胸腺细胞增殖从而阻止新鲜T细胞的补充。  相似文献

Summary The effects of selenium on T lymphocyte proliferation and its mechanisms have been investigated. The results showed that selenium is able to enhance lectin-stimulated T lymphocyte proliferation, to increase the production of interleukin 2 (IL-2) by lymphocytes and interleukin 1 (IL-l) by macrophages in the presence of lectin and to augment the response of T lymphoblasts to IL-2 and that of thymocytes to IL-1. The data presented suggest that selenium may be an important modulator for immune response. Selenium might enhance IL-2 production and response through the augmentation of IL-1 production and response, thereby promoting the enhancement of the proliferation and action of T lymphocytes and other immunocompetent cells. Based on these results, we may consider using selenium as an immunological enhancement agent to enhance or recover immune functions of the organism.  相似文献

目的:研究苦参在体内对免疫功能的作用。方法:小鼠口喂苦参溶液2周后检测脾细胞和胸腺细胞对ConA和LPS刺激的增殖反应,脾细胞产生白细胞介素2(IL-2),以及腹腔巨噬细胞产生白细胞介素1(IL-1)等指标。结果:口服苦参组上述三项指标均明显低于饮用自来水的对照组。结论:苦参在体内对免疫功能具有很明显的抑制作用。  相似文献

In this report the authors studied the effects of kang shuai sen fang (KSSF) on the immune functions of mice. The results indicated that KSSF was able to enhance the proliferation response of spleen cells of mice to Con A and LPS. At the dosage of 400 mg/kg.d, KSSF could stimulate the production of plague forming cells (PFC), the DTH response induced by allogeneic splenocytes. The mixed lymphocyte reaction was much stronger in the drug-receiving group than that in the control group. Further study indicated that KSSF was able to enhance the activities of cytotoxic T lymphocyte and the production of interleukin 2.  相似文献

目的:观察乙酰胆碱(ACh)对大鼠淋巴细胞由刀豆素A(ConA)诱导的白细胞介素2(IL-2)生成的影响。方法:取大鼠的脾脏制成单个细胞悬液进行体外培养,用ConA诱导脾细胞的IL-2生成,然后以MTT比色法间接测定IL-2的生成。结果:ACh,M型胆碱能受体激动剂毛果芸香碱和N型胆碱能受体激动剂烟碱在10^-10-10^-6mol/L浓度范围都能显著增强ConA诱导的IL-2生成,M型胆碱能受体阻断剂阿托品(10^-10和10^-9mol/L)能阻断同浓度ACh的增强作用;N型胆碱能受体阻断剂筒箭毒碱(10^-10和10^-9mol/L)可部分阻断同浓度ACh的增强作用。结论:在机体内副交感神经兴奋可导致T淋巴细胞的功能增强,其免疫调节作用由免疫细胞上的M和N受体介导,可能M受体的作用占主导地位。  相似文献

研究西洋参多糖(polysaccharidefrompanaxquinquefoliumPPQ)对小鼠脾淋巴细胞刺激效应。通过3H掺入法测淋巴细胞转化及母细胞增殖法测IL-2合成。结果表明,PPQ可协同亚剂量ConA促进脾淋巴细胞转化及IL-2合成。将PPQ—1进一步分离得到4种成分(PPQ-1-1~4)。体外实验结果表明,PPQ-1-1~4既可单独也可协同亚剂量ConA促进脾淋巴细胞转化。提示西洋参多糖对小鼠脾淋巴细胞具有一定刺激作用。  相似文献

Summary In this paper, we report our work about the effect of insulin on the expression of interleukin 2 receptor (IL-2R). Our experiments showed that insulin was not T cell growth factor, but was able to augment the expression of IL-2R during T cell activation by PHA and to delay the decline rate of IL-2R+ cell. As a result, the percentage of IL-2R+ cells significantly increased during and after activation. Furthermore, insulin was capable of enhancing the production of interleukin 1 by macrophage and interleukin 2 by T lymphocyte. Based on these results, we suggest that there are at least two related mechanisms involved in the augmentation of the expression of IL-2R on activated T cells, i.e., direct action of insulin on T cells and indirect action of insulin by enhancing the production of interleukin l.  相似文献

本实验研究了创伤小鼠巨噬细胞对T淋巴细胞的直接接触抑制作用。结果显示:创伤后巨噬细胞可通过直接的细胞接触方式抑制正常小鼠T淋巴细胞转化、IL-2的产生、IL-24的表达以及IL-2介导的淋巴细胞增殖,但对IL-2-IL-2R反应及IL-1的产生无明显影响,可明显提高Ts细胞抑制活性。去除正常小鼠淋巴细胞中的Ts细胞,可阻断创伤后巨噬细胞的这一抑制活性。提示:(1)创伤后巨噬细胞可通过IL-1以外的  相似文献

采用杞芪合剂治疗7/8肾切除制备的大鼠肾功能不全模型。结果发现杞芪合剂可明显降低大鼠血肌酐、甘油三酯及尿蛋白含量,抑制巨噬细胞产生IL-1,促进淋巴细胞产生IL-2,改善肾衰大鼠的贫血状况,减轻其病理改变,提示杞芪合剂对慢性肾衰大鼠有明显的治疗作用。  相似文献

目的:探讨中药内异方对子宫内膜异位症大鼠细胞因子含量及血管内皮细胞生长因子(VEGF)mRNA表达的调节作用。方法:用Cummings AM方法复制子宫内膜异位症动物模型,采用ELISA法检测IL-6、IL-8、IL-10含量,Dot blot法检测VEGF mRNA表达水平。结果:模型组外周血IL-6、腹腔液IL-8水平、腹腔巨噬细胞数量及巨噬细胞在脂多糖(LPS)刺激下产生的IL-6含量显高于正常对照组;内异方组异位种植内膜组腹腔巨噬细胞在LPS刺激下产生的IL-10含量显低于正常对照组和内异方组;模型组种植异位内膜组织VEGF mRNA表达水平明显高于内异方组。结论:子宫内膜异位症细胞因子IL-6、IL-8、IL-10的失衡与疾病的发生发展有关,VEGF mRNA的过度表达与内异灶血管的形成相关;内异方可显抑制异位内膜组织的生长,同时抑制异位灶VEGF mRNA的过度表达,减少异位灶血管的生成,并对细胞因子有明显的调节作用。  相似文献

目的 探讨内脏脂肪素是否能调节内皮细胞单核细胞趋化蛋白1(MCP-1)和白介素6(IL-6)生成以及胰岛素受体(IR)的介导作用.方法 不同剂量和不同干预时间的内脏脂肪素刺激3~5代脐静脉内皮细胞(HUVEC);然后在内脏脂肪素(100 ng/m1)刺激基础上加入IR酪氨酸激酶抑制剂预处理HUVEC,24 h后测定MCP-1和IL-6蛋白和基因表达.酶联免疫吸附法和实时定量聚合酶链反应检测MCP-1和IL-6蛋白和mRNA表达.结果 内脏脂肪素剂量和时间依赖性促进HUVEC合成MCP-1[剂量效应:对照组:(62±10) pg/ml、100 ng/ml:(273±65) pg/ml、500 ng/ml:(1630±103) pg/ml;时间效应:对照组:(37±27) pg/ml、12 h:(184±56) pg/ml、48 h:(328±80) pg/ml]和IL-6[剂量效应:对照组:(31.8±1.7) pg/ml、100 ng/ml:(42.5±5.7) Pg/ml、500 ng/ml:(154.7±10.2) pg/ml;时间效应:对照组:(15.7±3.4) pg/ml、12 h:(35.4±4.7) pg/ml、48 h:(77.6±11.8) pg/ml].IR酩氨酸激酶抑制剂抑制内脏脂肪素诱导的MCP和IL-6蛋白和基因表达.结论 内脏脂肪索能诱导HUVEC合成和分泌MCP-1、IL-6,其作用通过IR信号通路介导.  相似文献

目的:观察低剂量X射线照射对小鼠肝癌H-22细胞膜相关抗原肽(TAP)提取物抑瘤作用的影响,为低剂量辐射与肿瘤疫苗联合治疗肿瘤提供实验依据。 方法:采用微酸洗脱法制备分子量≤3 000的细胞膜TAP提取物,皮下免疫小鼠,免疫前12 h给予75 mGy X射线全身照射,检测胸腺细胞周期时相、脾细胞Con A反应性及脾T细胞CD4、CD8亚组的变化,同时观察体内抑瘤效应。 结果:TAP提取物免疫小鼠后,移植肿瘤的发生率降低,平均出现时间延迟,生长速度减慢;脾细胞Con A反应性增强,胸腺细胞周期百分数无显著变化。小鼠免疫前12 h给予75 mGy全身照射可增强TAP提取物的抑瘤效应,与单纯TAP组相比,胸腺细胞S期百分数明显增加,脾细胞CD8+ T细胞百分数增高。 结论:低剂量辐射能进一步激发免疫系统功能,增强小鼠肝癌H-22细胞膜TAP提取物的抑制肿瘤作用。  相似文献

OBJECTIVE: To investigate the effects of an intradermal injection of oligodeoxynucleotides (ODNs) containing unmethylated CpG motifs on concanavalin A (Con A)-induced hepatitis, an experimental model of immune-mediated hepatitis. METHODS: Con A was injected intravenously into Balb/c mice. Twelve hours after Con A challenge, blood and liver samples were obtained. CpG ODN was injected intradermally 48 hours before Con A challenge. The extent of liver injury was assessed by determining serum alanine transaminase (ALT) and by liver histology. Serum levels of cytokines, including interferon (IFN)-gamma, tumor necrosis factor (TNF)-alpha, interleukin (IL)-4 and IL-5, were measured by enzyme-linked immunosorbent assay. RESULTS: Co-administration of Con A and CpG ODN significantly increased serum ALT in mice compared with that in the case of administration of Con A alone (10,268 +/- 4,654 and 1,140 +/- 832 IU/1, respectively, p<0.05). In liver histology, mice treated with CpG ODN and Con A showed more extensive midzonal necrosis than did mice treated with Con A alone. These mice also showed significant increases in serum TNF-alpha and IFN-gamma and decrease in serum IL-5. CONCLUSIONS: The results indicate that CpG ODNs aggravate Con A-induced hepatitis by stimulating the production of T-helper-1 (Th1) cytokines, TNF-alpha and IFN-gamma, suggesting that bacterial DNA that contains unmethylated CpG motifs may contribute to the exacerbation of immune-mediated liver injury.  相似文献

接种肉瘤180,艾氏腹水癌和路易氏肺癌的小鼠,喂饲嗜酸性乳酸杆菌(Shahani 株)发酵乳后,小鼠肉瘤180和艾氏腹水癌生长受抑制。与对照组比较.差异明显(P<0.05).但它对路易氏肺癌自发性肺转移无影响。此外,发现 C_(57)BL/6小鼠喂饲该细菌发酵乳后,脾细胞和胸腺细胞对 ConA 刺激的增殖反应,腹腔巨噬细胞 C_(3b)受体活性以及脾细胞和腹腔渗出细胞所介导的对 L_979纤维母细胞瘤细胞的毒性作用都比对照组明显增强。  相似文献

Summary Anti-inflammatory Agent No.6 for injection (AI-6) is an effective anti-infective agent composed of four traditional Chinese medicinal herbs. It was demonstrated in our experiment that the agent was able to enhance oxidative burst of rat peritoneal macrophages as shown by the increased production of H2O2 and increased reduction of nitroblue tetrazolium (NBT). It seems likely that by these means AI-6 could promote the bactericidal function of macrophages. NBT reduction of macrophages was observed by both spectrophotometric quantification and electron microscopy. Project supported by the Science Fund of the Chinese Academy of Sciences.  相似文献

目的研究玉竹提取物A(extraction A of polygonatum odoratum,EA-PAOA)对小鼠MΦ细胞因子产生的影响,以探讨对免疫功能的作用。方法检测小鼠体外MΦ产生的IL-1和TNF-α。结果10ug/ml以上浓度的玉竹提取物A对小鼠MΦ产生的IL-1有抑制作用,1000ug/ml浓度的玉竹提取物A的IP(inhibitory perentage)达39.1%,对TNF-α抑制作用比对IL-1的抑制作用明显,10ug/ml浓度的玉竹提取物A的CIP(ytolytic-Inhibitory perentage)达28.6%,1000ug/ml浓度的玉竹提取物A的CIP达78.6%。两者均呈量效依赖关系。结论玉竹提取物A对小鼠MΦ产生的IL-1和TNF-α均有抑制作用,可抑制小鼠的免疫功能。  相似文献
