Streptococcus mutans is one of the major pathogens of dental caries. Oxyresveratrol, a natural compound found in plants, exerts inhibitory effects on many bacterial species but its effect on S. mutans is unknown. The objective of this study was to clarify the antibacterial effect of oxyresveratrol on S. mutans, including effects on basic viability, acidogenicity, acidurity, and extracellular polysaccharide synthesis. The expression of nine genes that encode virulence and protective factors in S. mutans was measured by qRT-PCR. Oxyresveratrol showed a dose-dependent inhibitory effect on survival of S. mutans. At 250 μg ml−1, oxyresveratrol reduced the S. mutans survival rate, inhibited synthesis of water-insoluble glucans, compromised biofilm formation, and significantly down-regulated the expression of glucosyltransferase-I (gtfB) and glucosyltransferase-SI (gtfC). However, the enzymatic activity of lactate dehydrogenase protein was increased and the expression of lactate dehydrogenase (ldh) and ATP synthase subunit beta (atpD) genes were also up-regulated. Besides, glucosyltransferase S (gtfD) up-regulation indicated that water-soluble glucan synthesis was promoted. The vicR, liaR, and comDE genes, which exert a self-protective function in response to external stress, were also up-regulated. In conclusion, oxyresveratrol inhibited the growth of S. mutans and also reduced biofilm formation, acid production, and synthesis of water-insoluble glucans by this organism. In addition, oxyresveratrol also activated a series of S. mutans self-protection mechanisms.  相似文献

The purpose of this study was to assess the success of the dental hygienist in the dental health education of dental patients in general dental practice. Special emphasis was laid on health education in the field of periodontics. The answers on a questionnaire of 159 dental patients who visited a dental hygienist were compared with the answers of 302 dental patients who had never visited a dental hygienist. The comparison was done by means of t-test and regression analysis on specially developed scales with regard to knowledge, motivation, self-care, and perception of change. On all scales the dental patients who visited a dental hygienist scored better. This difference remained when social demographic variables and the dentist's influence were taken into account. This indicates that the dental hygienist is successful in the behavioral aspect of her work.  相似文献

Comparison of coconut water, propolis, HBSS, and milk on PDL cell survival   总被引:3,自引:0,他引:3  
Coconut water is biologically pure and sterile, with a rich presence of amino acids, proteins, vitamins, and minerals. The purpose of this study was to use a collagenase-dispase assay to investigate the potential of a new storage medium, coconut water, in comparison with propolis, Hank's balanced salt solution (HBSS), and milk in maintaining viable periodontal ligament (PDL) cells on simulated avulsed teeth. Seventy freshly extracted human teeth were divided into 4 experimental groups and 2 control groups. The positive and negative controls corresponded to 0-minute and 8-hour dry times, respectively. The experimental teeth were stored dry for 30 minutes and then immersed in 1 of the 4 media (coconut water, propolis, HBSS, and milk). The teeth were then treated with dispase grade II and collagenase for 30 minutes. The number of viable PDL cells was counted with a hemocytometer and analyzed. Statistical analysis showed that coconut water kept significantly more PDL cells viable compared with propolis, HBSS, or milk. Coconut water can be used as a superior transport medium for avulsed teeth.  相似文献

OBJECTIVE: This study examined the effects of elevated-concentration hydrogen peroxide tooth whitening strips on the surface and subsurface integrity of enamel, coronal dentin, and root dentin. METHODOLOGY: Sound human teeth were ground and polished to prepare a uniform substrate for bleaching. A cycling regimen included exposures to saliva, bleaching treatments, and fluoridated toothpaste. Bleaching was carried out with plastic strips containing hydrogen peroxide gel at 11.7 and 14% concentrations. A non-bleached group served as a control. Bleaching times measured up to 45 hours. Physical properties of tooth surfaces were assessed by microhardness, while surface morphological properties of teeth were examined by profilometry, surface Confocal Laser Scanning Microscopy (CLSM), and Variable Pressure Scanning Electron Microscopy (VP-SEM). Subsurface histomorphological effects on teeth were assessed by CLSM. Lastly, the influences of bleaching on tooth micro-chemical composition was studied by micro-Raman spectroscopy using a unique spectrometer in line, coupled to the CLSM via glass fiber. RESULTS: Surface microhardness, profilometry, CLSM, and VP-SEM measures showed enamel and root dentin surfaces were unchanged with bleaching. CLSM micromorphological assessments demonstrated normal histology for bleached teeth on surface and subsurface enamel, DEJ, and dentin. Raman spectroscopy demonstrated no effects of bleaching treatments on the microchemical mineral composition of enamel and dentin. Bleaching treatments were observed to reduce background luminescence of enamel, DEJ, and dentin. CONCLUSION: These results confirm that whitening strips delivering controlled doses of hydrogen peroxide at 11.7 and 14% concentrations do not produce changes in surface/subsurface histomorphology, surface microhardness, or micro-chemical mineral composition of teeth. The effects of bleaches on tooth luminescence recorded in micro-Raman spectroscopy may serve as an internal signature to bleaching effects and warrant further study.  相似文献
