目的:探讨临产前、后子宫平滑肌细胞间隙连接蛋白Cx-43的表达以及母血、脐血、羊水中促肾上腺皮质激素释放激素(CRH)、皮质醇(Cortisol)的水平变化,分析它们与分娩发动的关系。方法:应用免疫组化SABC法结合计算机图象分析技术分析60例足月妊娠产妇子宫下段平滑肌Cx-43的表达;用放射免疫方法测定产妇的静脉血、羊水及胎儿脐静脉血中CRH、Cortisol的含量。结果:临产组子宫肌细胞Cx-43的表达明显强于未临产组,差异有显著性(P<0.01)。临产组母血、脐血中CRH、Cortisol及羊水中Cor-tisol与临产前比较显著升高(P<0.01),且与子宫肌Cx-43蛋白的表达呈显著正相关(P<0.01);结论:CRH、Cortisol在分娩发动中起重要作用,并可能通过促进子宫平滑肌Cx-43的表达参与分娩发动。  相似文献

目的 探讨子宫平滑肌组织中RhoA及Rho激酶的表达变化及其在分娩发动中的作用.方法 选择2007年2至12月在中南大学湘雅二医院妇产科住院分娩的足月临产产妇20例(临产组),足月未临产产妇20例(未临产组);同期因宫颈上皮内瘤变行子宫切除术的患者20例为对照组.应用实时荧光定量RT-PCR技术和蛋白印迹法检测3组妇女子宫平滑肌组织中RhoA和Rho激酶亚型ROCKⅠ mRNA及其蛋白的表达水平,并对子宫平滑肌组织中RhoA mRNA及蛋白与ROCKⅠ mRNA及蛋白的表达水平行相关性分析.结果 (1) 3组妇女子宫平滑肌组织中RhoA mRNA和ROCKⅠmRNA的表达水平,临产组分别为(3.51±0.56)×10-3和(4.07±0.66)×10-3;未临产组分别为(2.75±0.52)×10-3和(2.71±0.52)×10-3;对照组分别为(2.11±0.54)×10-3和(2.01±0.23)×10-3.临产组产妇子宫平滑肌组织中RhoA mRNA和ROCKⅠ mRNA的表达水平均高于未临产组和对照组, 差异有统计学意义(P<0.01);3组间分别比较,差异均有统计学意义(P<0.05).(2)3 组妇女子宫平滑肌组织中RhoA蛋白和ROCKⅠ蛋白的表达水平,临产组分别为0.72±0.23和0.56±0.14;未临产组分别为0.64±0.17和0.42±0.16;对照组分别为0.46±0.15和0.29±0.08.RhoA蛋白在3组中的表达两两比较,差异均无统计学意义(P>0.05).临产组和未临产组产妇子宫平滑肌组织中ROCKⅠ蛋白表达水平均高于对照组,差异均有统计学意义(P<0.01).(3)3组妇女子宫平滑肌组织中RhoA mRNA表达与ROCKⅠ mRNA的表达水平呈正相关关系(r=0.73,P<0.01);3组妇女子宫平滑肌组织中RhoA蛋白表达与ROCKⅠ蛋白表达水平也呈正相关关系(r=0.37,P<0.01).结论 子宫平滑肌组织中RhoA及Rho激酶水平升高与产妇分娩发动密切相关.  相似文献

Methods Forty term pregnant women, who delivered through cesarean section at the Xiangya Second Hospital of Central South University from February 2007 to November 2007, were selected and divided into 2 groups: 20 in labor group and 20 in non-labor group. Another 20 non-pregnant women undergoing hysterectomy due to cervical intraepithelial neoplasia were chosen as the control. Real-time fluorescence quantitative RT-PCR and western blot were applied to detect the expression RhoA and ROCKⅠ mRNA and protein in uterine smooth muscle tissue and the correlation between the mRNA and protein expression of RhoA and ROCKⅠ were analyzed. Results (1) The mRNA expressions of both RhoA and ROCKⅠ were detected in all groups, and higher levels were found in the labor group than in the non-labor group and the control [RhoA mRNA: (3.51±0.56)×10-3 vs. (2.75±0.52)×10-3 and (2.11±0.54)×10-3; ROCKⅠ mRNA: (4.07±0.66)×10-3 vs. (2.71±0.52)×10-3 and(2.01±0.23)×10-3, P<0.01]. (2) RhoA and ROCKⅠ proteins were also identified in all three groups, and the expressions in the labor and non-labor group were higher than those of the control (RhoA protein: 0.72±0.23 and 0.64±0.17 vs. 0.46±0.15; ROCKⅠ protein: 0.56±0.14 and 0.42±0.16 vs. 0.29±0.08, P<0.01). (3) The expression of RhoA mRNA and ROCKⅠ mRNA were positively correlated in each of the three groups (r=0.73,P<0.01), and the same was found in the expression of RhoA protein and ROCKⅠ protein (r=0.37,P<0.01). 目的 探讨子宫平滑肌和胎膜组织表皮生长因子受体 (EGFR)mRNA表达水平与分娩发动的关系。方法 用半定量逆转录聚合酶链反应检测分娩发动前、后各 10例产妇子宫平滑肌和胎膜组织的EGFRmRNA表达水平。结果 子宫平滑肌EGFRmRNA表达水平由分娩发动前的 0 .91± 0 .3 2增高至分娩发动后的 1.3 3± 0 .2 3 ,两者比较 ,差异有显著性 (P <0 .0 5 ) ;胎膜组织EGFRmRNA表达水平由分娩发动前的 1.0 5± 0 .3 0增高至分娩发动后的 1.65± 0 .71,两者比较 ,差异有显著性 (P<0 .0 5 )。结论 分娩发动前、后子宫平滑肌及胎膜组织中EGFRmRNA表达水平升高 ;并与分娩发动有密切关系 ,是诱发子宫平滑肌收缩的重要因素。  相似文献

目的 探讨子宫结合带的超微结构特点及间隙连接蛋白43(Cx43)在子宫结合带中的表达变化与子宫腺肌病发病的关系.方法 收集2008年11月至2009年11月在内蒙古医学院附属医院妇科住院的因子宫腺肌病行子宫全或次全切除术者30例为观察组(其中子宫内膜为增生期14例,分泌期16例),选择同期因宫颈上皮内瘤变(CIN)Ⅲ等行子宫全切除术者30例为对照组(子宫内膜为增生期、分泌期各15例).采用链霉素亲和素-生物素-过氧化物酶复合物(SABC)免疫组化法检测Cx43蛋白在子宫腺肌病患者子宫结合带中的表达水平,以吸光度(A)值表示;透视电镜观察两组中无诊断性刮宫(诊刮)史、剖宫产史、子宫手术史者(各4例)的子宫在位内膜、子宫结合带以及子宫肌层外1/3的超微结构特点.结果 (1)Cx43蛋白的表达:观察组增生期及分泌期子宫结合带中Cx43蛋白表达水平分别为0.133±0.018、0.137±0.021,对照组分别为0.154±0.016、0.141±0.018,两组同一期别子宫内膜比较,差异均有统计学意义(P<0.05),但各组内增生期与分泌期子宫结合带中Cx43蛋白表达水平比较,差异无统计学意义(P>0.05);观察组的在位内膜中增生期及分泌期Cx43蛋白表达水平(0.067±0.017,0.062±0142)低于对照组(0.094±0.005,0.080±0.005),差异也均有统计学意义(P<0.05),但各组内增生期与分泌期比较,差异无统计学意义(P<0.05);观察组和对照组增生期外肌层Cx43蛋白表达分别为0.184±0.022、0.188±0.028,差异无统计学意义(P>0.05),分泌期外肌层Cx43蛋白表达分别为0.178±0.022、0.191±0.025,差异也无统计学意义(P>0.05).(2)形态学变化:观察组子宫结合带平滑肌细胞核面积[(24.3±1.6)μm2]比对照组[(21.8±2.0)μm2]明显增加,差异有统计学意义(P<0.01).细胞膜致密斑明显增长[(1.07±0.17)、(0.71±0.07)μm],差异也有统计学意义(P<0.01);观察组子宫结合带肌细胞小凹结构减少,细胞增生肥大,排列紊乱,核表面呈圆齿状不规则外形,核染色质边集,异染色质多,细肌丝较少而中间丝较多,线粒体数量增加、体积增大、空泡化明显,粗面内质网扩张,高尔基体增加,肥大细胞与肌细胞紧密相邻;在位内膜的腺上皮细胞间呈桥粒连接,且表面绒毛增粗、致密.以上特点在观察组的外肌层也可见,但没有子宫结合带显著.结论 子宫结合带中超微结构的变化和Cx43蛋白的表达下调可能与子宫腺肌病的发病有关.  相似文献

目的 :研究间隙连接蛋白 4 3(Cx4 3)在子宫肌瘤中的表达及米非司酮作用后Cx4 3蛋白表达的改变 ,探讨子宫肌瘤的生长及治疗的相关机制。方法 :用免疫组化法检测 30例子宫肌瘤组织 ,2 0例术前 3d给予米非司酮的子宫肌瘤及 2 0例正常子宫肌组织中Cx4 3蛋白的表达。结果 :子宫肌瘤Cx4 3蛋白表达高于正常子宫肌组织 (P <0 .0 5 )。子宫肌瘤Cx4 3蛋白表达增生期高于分泌期 (P <0 .0 5 )。米非司酮作用后Cx4 3蛋白表达在增生期无明显改变 (P >0 .0 5 ) ,分泌期明显增高 (P <0 .0 5 )。结论 :子宫肌瘤中Cx4 3蛋白表达水平与月经周期相关 ,米非司酮对Cx4 3蛋白表达的上调可能是其治疗子宫肌瘤的机制之一。  相似文献
