目的研究不同浓度的染料木黄酮(genistein,GEN)对体外培养大鼠成骨细胞游离钙、钙调素、碱性磷酸酶(alkaline phosphatase,ALP)、ATP酶的影响。方法采用新生大鼠颅骨分离培养成骨细胞,加入不同浓度的染料木黄酮(10^-8mol/L,10^-7mol/L,10^-6mol/L,10^-5mol/L),以妊马雌酮(商品名:倍美力)(10^-8mol/L,10^-7mol/L,10^-6mol/L,10^-5mol/L)作为阳性对照组,用PNPP(对硝基苯磷酸盐)法测定ALP活性、放射生物法测定钙调素水平、激光扫描共聚焦显微镜测定细胞游离钙、比色法测定ATP酶活性。结果染料木黄酮对成骨细胞钙调素、ALP、Ca^2+-ATP、细胞内游离钙水平的影响呈正相关性,并与其剂量成正相关性,但这些作用均低于雌激素水平(P〈0.01)。结论染料木黄酮能增加成骨细胞ALP、Ca^2+-ATP活性和钙调素水平、钙离子震荡波,并与其浓度有关,作用与雌激素相似。  相似文献

目的探讨脱氢表雄酮(dehydroepiandrosterone,DHEA)对离体大鼠成骨细胞和IL-1β的影响。方法体外分离培养大鼠成骨细胞,取传一代细胞,分别给予10^-5mmoL/L、10^-7mmoL/L、10^-9mmol/LDHEA培养72h,以10^-8mmoL/L雌二醇(estradiol,E2)为阳性对照,另设空白对照组。碱性磷酸酶(alkaline phosphatase,ALP)染色鉴定成骨细胞,MTT法检测成骨细胞的增殖能力,PNPP法测定ALP活性。ELISA法测定不同浓度DHEA培养液中IL-1β的水平。结果不同浓度DHEA组成骨细胞增殖能力和ALP的活性均有上升,其中以10^-7mmoL/LDHEA组最显著(P〈0.01),其作用和E2相似(P〉0.05);10^-9mmoL/LDHEA组单位细胞数的ALP活性高于空白对照组(P〈0.05)。不同浓度的DHEA组IL-1β呈下降趋势,成骨细胞增殖能力及ALP活性和IL-1β成负相关。结论DHEA在体外促进大鼠成骨细胞生长和增殖,提高ALP活性,其作用可能和抑制IL-1β有关。  相似文献

目的探讨交感神经递质去甲肾上腺素对体外培养大鼠kupffer细胞TNF-α、IL-1β炎症相关细胞因子表达的影响。方法分离大鼠kupffer细胞体外分三组培养,经体外培养48h后,给予外源性内毒素(LPS10μg/m1)刺激,同时给予不同浓度的去甲。肾上腺素(0.1μmol/L~10μmol/L)分别作用12h,运用定量逆转录多聚酶链反应检测各处理组TNF-α和IL-1β mRNA的表达,ELISA检测细胞培养上清TNF-α IL-1β蛋白的表达。结果同时给予LPS(10μg/ml)刺激和去甲肾上腺素(1μmol/L及10μmol/L)作用后,kupffer细胞培养上清TNF-α和IL-1β蛋白较LPS组显著增高(P〈0.05);TNF-α mRNA表达分别较LPS组增加50.9%(P〈0.05)和59.1%(P〈0.05);IL-1β mRNA较LPS组表达增加53.7%(P〈0.05)和57.8%(P〈0.05)。结论应激浓度的去甲肾上腺素能增加内毒素诱导大鼠kupffer细胞TNF-α、IL-1β表达,具有促炎效应。  相似文献

目的研究体外辛伐他汀对钛颗粒刺激单核细胞形成破骨细胞的影响,探讨辛伐他汀防治人工关节无菌性松动的可能性。方法体外分离培养人外周血单个核细胞并分成5组,A组为钛颗粒刺激组(单核细胞和磨屑混合培养),B组为10^-5mol/L辛伐他汀组(单核细胞、磨屑混合培养+10^-5mol/L辛伐他汀),C组为10^-6mol/L辛伐他汀组(单核细胞、磨屑混合培养+10^-6mol/L辛伐他汀),D组为10^-7mol/L辛伐他汀组(单核细胞、磨屑混合培养+10^-7mol/L辛伐他汀),E组为单核细胞组。各组细胞培养24h后取上清液,用ELISA法检测上清液中肿瘤坏死因子(TNF-α)、单核细胞趋化蛋白-1(MCP-1)的含量。分别培养10d、18d后进行TRAP染色阳性细胞计数,采用扫描电镜检测骨磨片的吸收陷窝,观察钛颗粒对破骨细胞形成的影响。结果磨屑刺激单个核细胞分泌溶骨因子,辛伐他汀抑制磨损颗粒刺激单核/巨噬细胞分泌TNF-α及MCP-1;且破骨细胞数明显减少,骨吸收陷窝数减少,与钛颗粒组刺激组比较,差异均有统计学意义(P〈0.05)。结论辛伐他汀通过抑制TNF-α、MCP-1的释放而有效防止磨屑诱导的骨溶解,有望成为防治人工关节无菌性松动的一种有潜力的药物。  相似文献

目的:通过观察在不同浓度枸橼酸他莫昔芬处理下,大鼠骨髓基质干细胞(rBMSCs)诱导成脂后生长状态的变化,初步探讨用于促进骨髓基质干细胞诱导成脂的最适枸橼酸他莫西芬浓度。方法:培养大鼠骨髓基质千细胞并传代,将第三代的骨髓基质干细胞随机分为三组,分别在加入10^-6mol/L,10^-7mol/L,10^-8mol/L的枸橼酸他莫西芬24h后,进行成脂肪的诱导。分别对三组样本进行四甲基偶氮唑盐检测(MTT检测),测量其光密度值(optical density,OD值)。取第8天的样本进行油红-0染色,计算红色脂滴的数量,并通过多个样本均数的方差分析比较三组之间的差别。结果:①各组OD值之间的差异没有统计学意义(P〉0.05):②10^-6mol/L组成脂数量明显多于10^-7mol/L组和10^8mol/L组,P值均小于0.01;而10^-7mol/L和10^-8mol/L两组之间在成脂数量上的差异没有显著统计学意义(P〉0.05)。结论:10^-6mol/L为枸橼酸他莫西芬促进大鼠骨髓基质干细胞诱导成脂的适宜浓度。  相似文献

[目的]检测依那西普(Etanercept,Enbrel)对钛颗粒刺激巨噬细胞(macrophages,MФ)分泌肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)、白介素1(interleukin-1,IL-1)、白介素6(interleukin-6,IL-6)的影响,探讨依那西普防治人工关节无菌性松动的可能性。[方法]分离、培养小鼠腹腔巨噬细胞,24h后分为5组。A组:仅为MФ;B组:MФ+钛颗粒;C组:MФ+钛颗粒+依那西普(10ng/m1);D组:MФ+钛颗粒+依那西普(100ng/ml);E组:MФ+钛颗粒+依那西普(1000ng/ml)。培养18h后,用酶联免疫法(ELISA)检测细胞培养上清液中TNF-α、IL-1、IL-6的含量。[结果]B组TNF-α、IL-1、IL-6含量明显高于A、D、E组(P〈0.001),E组TNF-α、IL-1、IL-6的含量明显低于B组、C组(P〈0.001,E组和D组间无统计学意义(P〉0.05)。[结论]钛颗粒可以刺激MФ分泌大量的TNF-α、IL-1、IL-6,依那西普能够呈剂量依赖型地有效抑制钛颗粒诱导的MФ分泌TNF-α、IL-1、IL-6,有望成为预防人工关节无菌性松动的药物。  相似文献

目的 初步探讨多种免疫抑制剂对嗜铬细胞瘤12(pheochromocytoma 12,PC12)细胞和L929细胞增殖的影响。方法 对数生长期PC12细胞和L929细胞传代,取细胞株复苏后第3代细胞均以1×10^6/ml密度接种于培养板中,分别加入10、10、10^-7和10^8mol/L环孢菌素A(cyclosporin A,CsA),10^-6、10^-7、10^-8和10^-9mol/LFK506以及10^-3、10^-4、10^-6和10^-8mol/L甲基强地松龙,并设立空白对照组。于培养24、48和72h后,取各浓度药物作用的细胞,采用MTT法检测细胞增殖。结果 高浓度(10mol/L)甲基强地松龙和较低浓度(10^-8~10^-7mol/L)CsA,在给药后48h内对PCI2细胞增殖有明显促进作用,此后促增殖作用不明显;而各个浓度的FK506均无促进PCI2细胞增殖的作用。高浓度甲基强地松龙(10^-3mol/L)和CsA(10^-6~10mol/L)作用24h后,对L929细胞增殖有显著抑制作用,FK506仅在较高浓度(10^-6mol/L)有一过性(仅出现于给药后48h)促进L929细胞增殖的作用。结论 10^-3mol/L甲基强地松龙和10^-8~10^-7mol/LCsA能够在短时间内促进PC12细胞增殖,而10^-3mol/L甲基强地松龙和10^-6~10^-5mol/L CsA对L929细胞增殖有显著抑制作用。  相似文献

目的:研究甲状旁腺激素(PTH)对大鼠系膜细胞合成与分泌转化生长因子β1(TGF-β1)的影响。方法:(1)分别以10^-12,10^-11,10^-10,10^-9,10^-8mol/l的hPTH1-34刺激大鼠系膜细胞6,12,24,48h后,ELISA方法测定上清中TGF-β1的浓度;(2)分别以10^-12,10^-11,10^-10,10^-9,10^-8mol/L的hPTH1-34刺激大鼠系细胞48h,采用半定量RT-PCR方法检测细胞TGF-β1 mRNA的表达;(3)以10^-8mol/L的hPTH1-34分别刺激大鼠系膜细胞6,12,24,48h,采用半定量RT-PCR方法检测细胞TGF-β1 mRNA的表达,结果:(1)ELISA结果显示,hPTH1-34促进大鼠系膜细胞合成与分泌TGF-β1分泌作用达高峰(P<0.01),(2)半定量RT-PCR方法结果显示,hPTH1-34促进大鼠系膜细胞TGF-β1 mRNA的表达,并具有浓度依赖和时间依赖性特点(各组与对照组间P<0.05),结论:hPTH1-34从蛋白和基因水平显著促进系膜细胞TGF-β1的合成与分泌,且呈浓度依赖和时间依赖性特点。  相似文献

目的探讨N-乙酰基-丝氨酰-天冬氨酰-赖氨酰-脯氨酸(AcSDKP)对真皮成纤维细胞表达TGF—β1的影响。方法对人皮肤成纤维细胞原代培养及鉴定后,给予不同浓度AcSDKP(10^-7mol/L、10^-8mol/L、10^-9mol/L、10^-10mol/L、10^-11mol/L)干预,设立空白对照组。WST-8法检测人皮肤成纤维细胞增殖;RT-PCR技术检测人皮肤成纤维细胞TGF~β1 mRNA表达:酶联免疫法检测培养细胞上清液TGF-β1含量。结果与空白对照组相比,各浓度组对成纤维细胞的增殖都有抑制作用,其中10^-10mol/LAcSDKP抑制成纤维细胞增殖作用最强;10^-7mol/L、10^-8mol/L、10^-9mol/L、10^-10mol/LAcSDKP对TGF-β1 mRNA表达有显著的抑制作用,且各浓度组间的抑制作用无差异;10^-8mol/L、10^-9mol/L、10^-10mol/LAcSDKP可使培养上清液中的TGF—β1显著减少,各浓度组间差异无显著性。结论AcSDKP能够抑制人皮肤成纤维细胞增殖及TGF—β1的生成,有望成为病理性瘢痕防治的新方法。  相似文献

目的 初步探讨多种免疫抑制剂对培养的大鼠巨噬细胞和雪旺细胞数量的影响。方法 取新生大鼠腹腔巨噬细胞和坐骨神经雪旺细胞进行培养,细胞生长稳定、定量计数后,分别加入不同浓度的环孢素A(ciclosporin A,CsA)、FK506和甲基强地松龙(CsA浓度为10^-5、10^-6、10^-7和10^-8mol/L;FK506浓度为10^-4、10^-7、10^-8和10^-9mol/L;甲基强地松龙浓度为10^-3、10^-4、10^-6和10^-8mol/L),设立不加药物的空白对照组。于培养24、48和72h后,分别取各浓度药物作用的细胞,用MTT法检测细胞数量。结果 对于巨噬细胞,甲基强地松龙仅在10^-4mol/L对其有明显保护作用,而CsA和FK506则分别在10^-6、10^-7、10^-8mol/L和10^-7、10^-8、10^-9mol/L能够较好地保护巨噬细胞。对于雪旺细胞,10^-3mol/L甲基强地松龙对其生长有显著抑制作用,而10^-8mol/L作用72h,则表现为对雪旺细胞明显的保护作用。除了10^-5mol/L CsA作用72h会显著减少培养的雪旺细胞数量外,其他浓度的CsA和FK506对雪旺细胞的数量无显著影响。结论 高浓度的甲基强地松龙以及一定浓度的CsA和FK506能够保护培养的巨噬细胞,但高浓度的甲基强地松龙和CsA可抑制雪旺细胞的生长,长时间、低剂量的甲基强地松龙对雪旺细胞有明显的保护作用。  相似文献

Background: Obesity affects the regulation of immune and inflammatory responses. This study characterizes differences in peripheral blood lymphocyte phenotype in obese humans. Methods: Frequencies of lymphocyte subsets among peripheral blood mononuclear cells were compared between 10 obese (BMI ≥35) and 10 lean subjects, as determined by antibodies directed against cluster differentiation (CD) markers. Results: Obese patients demonstrated an increased frequency of CD3+CD4+ T-cells (mean difference 12%, P=0.004), a decreased frequency of CD3+CD8+ T-cells (mean difference 9.4%, P=0.016) and an increased frequency of CD3+CD8+CD95+ T-cells (mean difference 13.3%, P=0.032). No other differences among T-cell or monocyte subsets were noted. Conclusions: Obesity is associated with alterations in frequencies of peripheral CD4+ and CD8+ T-cells and aberrations in the expression of CD95 among CD8+ T-cells. These data suggest both CD4+ and CD8+ T-cell compartments, as well as the regulation of CD95 expression on CD8+ T-cells, as targets for further study into obesity's effects on the immune system.  相似文献

对高海拔地区的27例烧伤病人动脉血气变化进行了分析和观察。结果证明:无论是存活病人还是死亡病人伤后均存在有低氧血症问题。并且在死亡病人和烧伤合并吸入性损伤病人其低氧血症的发生早于单纯烧伤病人。提示:吸入性损伤病人应立即行气管切开术以保障氧气供给,单纯烧伤病人可常规吸氧以维持正常血 PaO_2,ARDS 均发生在合并吸入性损伤的病人,高频喷射通气技术对纠正低氧血症有一定效果。  相似文献

目的 研究β—半乳糖苷酶(β—gal)在成骨细胞中的表达状况,为阐明MorquioB综合征的发病机制提供依据。方法 裸鼠各器官和骨组织标本行X-gal染色检测。抽取羊和人骨髓行骨髓基质细胞(BMSCs)培养,分为4组:I:Adv-hBMP-2转染组;Ⅱ:Adv—β—gal转染组;Ⅲ:未转染组;Ⅳ:地塞米松诱导组。分别行X-gal染色和RT-PCR检测β—gal的表达。结果 裸鼠骺板两侧、骨膜内面及松质骨的成骨细胞和破骨细胞可见多量β—gal的表达。未转染BMSCs组有少量β—gal的表达,其他3组细胞的β—gal表达增高。结论成骨细胞和破骨细胞可表达多量β—gal,该两种细胞的β—gal缺乏可能是MorquioB综合征骨骼异常的直接原因。  相似文献

IntroductionSmoking-attributable mortality (SAM) is a valuable indicator that can be used to characterize the course and health burden of the smoking epidemic. The aim of this paper was to estimate SAM in Spain in 2016 in the population aged 35 and over, using the best available evidence.MethodsA smoking prevalence-dependent analysis based on the estimation of population-attributable fractions was performed. Smoking prevalence (never, former, and current smokers) was calculated from a combination of the Spanish Health Survey (2016) and the European Health Survey (2014); the relative risk of death among current and former smokers was taken from the follow-up of various cohorts; and mortality rates were obtained from National Center for Statistics data. SAM estimates are presented globally, and by sex, age groups, and major disease categories: cancer, cardiometabolic diseases and respiratory diseases.ResultsIn 2016, 56,124 deaths were attributed to tobacco consumption, 84% in men (47,000), and 50% in the population aged over 74 (27,795). Overall, 50% of SAM was due to cancer (28,281), 65% of which was lung cancer. One in 4 attributable deaths (13,849) occurred before the age of 65.ConclusionsOne in 7 deaths in Spain in 2016 were attributable to smoking. This estimation of SAM clearly highlights the great impact of smoking on mortality in Spain, mainly due to lung cancer and chronic obstructive pulmonary disease.  相似文献

MicroRNAs(miRNAs or miRs) are small approximately 22 nucleotide RNA species that are believed to regulate diverse metabolic and physiological processes.In the recent past,several reports have surfaced that demonstrate the role of miRNAs in various biological processes and numerous disease states.For a disease as complex as diabetes,the emergence of miRNAs as key regulators leading to the disease phenotype has added a novel dimension to the area of diabetes research.On the other hand,the liver,a metabolic hub,contributes in a major way towards maintaining normal glucose levels in the body as it can both stimulate and inhibit hepatic glucose output.This equilibrium is frequently disturbed in diabetes and hence,the liver assumes special significance considering the correlation between altered hepatic physiology and diabetes.While the understanding of the mechanisms behind this altered hepatic behavior is not yet completely understood,recent reports on the status and role of miRNAs in the diabetic liver have further added to the complexities of the knowledge of hepatic pathophysiology in diabetes.Here,we bring together the various miRNAs that play a role in the altered hepatic behavior during diabetes.  相似文献

Ligated tubules from the corpus epididymidis of men and monkeys were incubated in medium containing horseradish peroxidase (HRP) as a marker for fluid-phase endocytosis. HRP was localized by light and electron microscopy after 0, 15, 30 and 60 min of incubation. Movement between the cells was prevented by tight junctions, but bypass of this barrier was apparently achieved by an intracellular vesicular mechanism leading to a time-dependent appearance of HRP in the lumen. Uptake of HRP into basal cells and capture by the lysosomal apparatus of principal cells were also observed. HRP-filled vesicles also appeared in the basal, mid and apical cytoplasm of epithelial cells in the caput 1 h after injection of the tracer into the epididymal circulation of the monkey, suggesting that this pathway also operates in vivo.  相似文献
