Using the model of acute infectious peritonitis in rats, it is shown that inflammation induced in the absence of mast cells is characterized by marked inhibition of reparative processes. The most significant accumulation of functionally active fibroblasts and the development of granulations and young connective tissue in the mesentery occur 5–10 days after flogogen injection in the natural development of inflammation and after 10–20 days in the absence of mast cells. The data suggest that under natural conditions mast cells directly or indirectly stimulate reparative processes. A factor inhibiting the proliferation of splenic colony-forming units, injected bothin vivo and after preincubation of mouse bone marrow cellsin vitro, had a dose-dependent effect on the increased proliferative activity of splenic colony-forming units from the bone marrow of mice treated with testosterone propionate. This was associated with a reduction in the number of early hemopoietic precursors of mouse bone marrow. The counts of clonogenic granulocytic-macrophagal and macrophagal colony-forming units decreased and that of burst-forming units in murine bone marrow increased after exposure to the hormone. Testosterone propionate promoted a decrease of the repopulating potential of bone marrow cells, which recovered after their preincubation with the factor inhibiting the proliferation of splenic colony-forming units. After separation of normal murine bone marrow cells in a Percoll density gradient cellular fractions with densities of 1.076 and 1.060 g/ml are capable of suppressing thein vitro growth of leukemia cells. The cytostatic activity of these fractions, however, does not surpass the level of antitumor antiproliferative activity intrinsic to intact bone marrow cells. These cells were found to be capable of joining the splenocytes, thymocytes, and lymph node cells in effector cytostatic cooperation and thus enhance the final antitumor effect. A radiometric method of evaluating the effect of chemical compounds on the proliferative activity of bone marrow cells is described which consists of measuring the incorporation of labeled3H-thymidine in DNA. Results are reported on a comparative study of the effect of known immunotropic substances on bone marrow cell proliferation using the present method and the method of evaluating endogenous colony formation. Analysis of the results obtained by two variants ofin vivo andin vitro experiments provides additional information regarding the mechanism of action of the substances. HLA levels were estimated in the plasma of allogeneic bone marrow recipients with and without graft-versus-host disease (GVHD). The presence of streptococcal M protein and A polysaccharide in culture medium is shown to have an inhibitory effect on the growth of clonogenic stromal precursors in cultures of healthy murine bone marrow and of healthy guinea pig bone marrow and spleen. The efficacy of colony formation dropped 1.5- to 2-fold in the presence of antigens in a concentration of 25 μg/ml in the medium. The inhibitory effect was absent if antigens were added to adhesive cell cultures. The addition of antigens to cultures originating from animals immunized with streptococcus resulted in inhibition of the efficacy of colony formation in complete cultures and in cultures of adhesive cells. The presence of streptococcal antigens in guinea pig stromal fibroblast cultures of different strains did not affect their growth or colony formation. These data indicate that the effects of streptococcal antigens appear to be aimed at the stromal cells not directly, but rather via another cellular category in the bone marrow and splenic cell cultures, probably lymphocytes. The effect of the purine receptor ligands N-ethylcarboxamide adenine and adenosine and of the purine antagonists mercaptopurine and azathioprine on the intracellular cAMP content in human peripheral blood lymphocytes and bone marrow lymphoblasts was studied. All preparations tested induced an increase in the cAMP level in peripheral blood lymphocytes. The selective immunosuppressive effect of adenosine antagonists may be due to their ability to modulate the activity of adenylate cyclase in lymphoid cells. The percent and absolute content of T helpers increases in the spleen of mice 4 and 17 h after resection of the liver or removal of a kidney, which is associated with an increase of the mitotic index of splenocytes. The number of T suppressors and the bone marrow cell mitotic index are unchanged. The T-helper/T-suppressor ratio in the spleen increases 1.7–2 times during surgery on the liver and kidney. The effect of biotechnologically obtained ubiquinone-10 on the level of photrin- and dioxydin-induced chromosomal aberrations is studied for different doses of the preparations by assessing chromosomal aberrations in bone marrow cells of C57B1/6 mice. Using the mouse model of acute infectious peritonitis caused byEscherichia coli, it is shown that the development of inflammation is accompanied by increases in the number of erythrokaryocytes, erythroid colony-forming units, and erythroid hematopoietic islets in the bone marrow and by rises in the activities of supernatants of cultured stimulated adherent and nonadherent myelokaryocytes and of peripheral blood. The results of this study indicate that a characteristic, feature of acute inflammation is strong activation of erythropoiesis with the development of hyperplasia of the erythroid marrow. The transfer of the human adenosine deaminase gene to murine stem hematopoietic cells is performed on an irradiated sublayer of a long-term bone marrow culture by the conventional method of retroviral transduction with cytokines and by stimulation of stem cells without cytokines. The efficiency of gene transfer into colony-forming units (CFUs) with the aid of cytokines is 72% and without them it is 50%. In irradiated mice reconstituted with the retrovirus-infected bone marrow cells the donor hematopoietic activity is preserved during a 1-year period. The proliferative activity of CFUs of chimeric cells 6 months after the reconstitution was the same and did not depend on the mode of gene transfer. The spleen repopulation activity is lowered in all the groups of chimeric mice 6–12 months after reconstitution. It is established that, during elongation of a segment of the extremity, normal regeneration of the bone tissue is attended by a slight decrease in monocyte adherence during the postoperative period and by an increase of the functional activity of lymphocytes during subsequent activation of osteogenesis.
