摘    要:目的:运用生物信息学方法探究GJB3基因在胰腺癌组织中的表达及潜在作用机制,并进一步验证其在胰腺癌组织中的表达水平及其临床意义。方法:利用GEPIA在线数据库分析GJB3在胰腺癌组织中的表达及对预后的影响;从TCGA数据库下载有关GJB3基因的临床样本表达数据,分析GJB3表达与胰腺癌临床病理参数的相关性;采用实时荧光定量PCR法测定15例胰腺癌组织及其癌旁组织中GJB3基因的表达水平,结合数据库临床病理参数分析其表达水平对胰腺癌患者预后的影响。随后通过慢病毒介导的RNA干扰来沉默AsPC-1细胞株中的GJB3表达,进一步检测胰腺癌细胞增殖能力及细胞周期所受到的影响。结果:GJB3在胰腺癌组织中的表达量高于正常胰腺组织(P<0.05)。与低表达患者相比,GJB3高表达患者的总体生存期(P<0.05)和无病生存期(P<0.05)较差。GJB3基因的表达与胰腺癌患者的T分期和TNM分期显著相关(P<0.05)。GJB3在胰腺癌患者的癌组织中表达水平明显增高(P<0.05)。沉默GJB3表达后,胰腺癌AsPC-1细胞的增殖能力受到抑制(P<0.05)。结论:GJB3基因在胰腺癌组织中的表达水平显著增高,与胰腺癌不良的预后相关,沉默GJB3能抑制胰腺癌细胞增殖,同时GJB3可能通过影响细胞周期加速胰腺癌的进展。

Abstract:Objective:Using bioinformatics methods to explore the expression and potential mechanism of GJB3 gene in pancreatic cancer (PaC) tissues,and to further verify its expression level in pancreatic cancer tissues and its clinical significance.Methods:The expression level of GJB3 mRNA in pancreatic cancer and its effect on prognosis were analyzed by GEPIA online database.To download the clinical sample expression data of GJB3 gene from TCGA database,and to analyze the correlation between GJB3 expression and clinical pathological parameters of pancreatic cancer.Real-time fluorescence quantitative PCR (RT-qPCR) was used to detect the expression level of GJB3 in 15 cases of pancreatic cancer and their paracancerous tissues (PC),combined with the clinical pathological parameters of the database,to analyze the effect of its expression level on the prognosis of pancreatic cancer patients.Then the expression of GJB3 in AsPC-1 cell line was silenced by lentivirus-mediated RNA interference to further detect the influence of cell proliferation and its cycle of pancreatic cancer cells.Results:The expression of GJB3 gene in pancreatic cancer tissues was significantly higher than that in normal tissues (P<0.05).Compared with patients with low expression,the overall survival (P<0.05) and disease-free survival (P<0.05) were poor in patients with high expression of GJB3.The expression of GJB3 gene was significantly correlated with T stage and TNM stage of patients with pancreatic cancer (P<0.05).The expression level of GJB3 in pancreatic cancer tissues was significantly higher than that in paracancerous tissues (P<0.05).After silencing GJB3 gene,the proliferation of pancreatic cancer AsPC-1 cells was inhibited,and silencing GJB3 gene can affect the progression of pancreatic cancer by inhibiting the cell cycle.Conclusion:The increased expression of GJB3 gene in pancreatic cancer is related to the poor prognosis.And silencing GJB3 can inhibit the proliferation of pancreatic cancer cells,and GJB3 may accelerate the progression of pancreatic cancer by affecting the cell cycle.
