EGR1在乳腺癌组织中的表达及临床意义

摘    要:目的:探究早期生长反应基因1(early growth response 1,EGR1)在乳腺癌组织中的表达及临床意义。方法:下载并提取TCGA数据库中EGR1的表达数据;购买乳腺癌组织芯片(HBreD090CS01),并采用免疫组织化学法检测EGR1在乳腺癌中的表达;利用Kaplan-Meier Plotter数据库分析EGR1在乳腺癌中的预后价值;利用TIMER数据库分析乳腺癌中EGR1表达与免疫细胞浸润的关系。结果:相较于癌旁组织,EGR1 mRNA在乳腺癌中的表达显著下调(P<0.001);EGR1蛋白在45例乳腺癌组织中有18例呈低表达,27例呈高表达,在45例癌旁组织中有9例呈低表达,36例呈高表达,差异具有统计学意义(P=0.038);EGR1表达与乳腺癌分化程度相关(P=0.003),与患者年龄、肿瘤大小、N分期、临床分期等无关(P>0.05);EGR1低表达的乳腺癌患者,其无复发生存率显著低于EGR1高表达的患者(P<0.001),但EGR1表达与乳腺癌患者的总生存率无明显相关性(P>0.05);乳腺癌组织中EGR1表达与B细胞浸润水平呈负相关(P<0.001),与CD8+T细胞、CD4+T细胞、巨噬细胞、中性粒细胞、树突状细胞浸润水平呈正相关(P<0.001)。结论:EGR1在乳腺癌组织中的表达较癌旁组织低,且与肿瘤分化程度、预后及免疫浸润有关,有望成为乳腺癌预后评估的分子标志物及治疗的潜在靶点。

Abstract:Objective:To explore the expression and clinical significance of early growth response 1(EGR1)in breast cancer.Methods:The expression data of EGR1 in the TCGA database was downloaded from the Xena website.The breast cancer tissue microarray(HBreD090CS01)was purchased from Outdo BioTech(Shanghai,China).Immunohistochemistry(IHC)staining was performed to detect the expression of EGR1 protein in breast cancer.The Kaplan-Meier Plotter database was used to analyze the prognostic value of EGR1 in breast cancer.The TIMER database was used to analyze the correlations between EGR1 expression and immune cell infiltration in breast cancer.Results: Compared with the adjacent tissues,the expression of EGR1 mRNA in breast cancer was significantly downregulated(P<0.001).In 45 breast cancer samples,18 samples showed low expression,and 27 cases samples showed high expression.In 45 para-tumor samples,9 samples showed low expression,and 36 samples showed high expression,the difference was statistically significant(P=0.038).EGR1 expression was correlated with the degree of tumor differentiation(P=0.003)and was not related to patient age,tumor size,N stage,and clinical stage(P>0.05).Besides,patients with low EGR1 expression hada lower recurrence-free survival rate than patients with high EGR1 expression(P<0.001),but EGR1 expression was not related to the overall survival rate of breast cancer patients(P>0.05).Furthermore,EGR1 expression in tumor tissues was negatively correlated with the level of B cell infiltration(P<0.001),and positively correlated with CD8+T cells,CD4+T cells,macrophages,neutrophils,and dendritic cells(P<0.001).Conclusion:The expression of EGR1 in breast cancer tissues is lower than that in adjacent tissues.Besides,EGR1 expression is related to tumor differentiation,prognosis,and immune cell infiltration,which is expected to be a novel biomarker for breast cancer prognostic assessment and a potential target for treatment.
