宫颈鳞状上皮癌变过程中水通道蛋白3的表达研究

摘    要:  目的  揭示水通道蛋白3(AQP3)在维吾尔族妇女宫颈癌发生、浸润过程中的表达及意义。  方法  应用RT-PCR、Western blot和免疫荧光染色检测AQP3在82例宫颈癌、36例宫颈上皮内瘤样病变Ⅲ级和42例正常宫颈中的表达。  结果  在正常宫颈和宫颈鳞癌组织中AQP3 mRNA和蛋白均表达,蛋白水平表达有差异(P < 0.05),而mRNA表达无差异(P>0.05)。AQP3在正常宫颈、宫颈上皮内瘤样病变Ⅲ级和宫颈癌中的阳性率分别为19.05%、27.78%和42.68%,阳性表达率存在显著性差异(P < 0.05)。结合临床病理资料分析,AQP3阳性表达率与宫颈癌患者临床分期、肿瘤直径、浸润深度呈正相关(P < 0.05),与宫颈癌大体分型、病理分级和淋巴结转移无关(P>0.05)。  结论  AQP3在子宫颈鳞癌疾病发生、肿瘤生长和局部浸润过程中可能发挥重要的生理作用。

Expression and Significance of Aquaporin 3(AQP 3) in Cervical Carcinogenesis

Abstract:  Objective  To reveal the expression and the clinical significance of aquaporin 3 (AQP 3) in cervical carcinoma tissues from Uygur patients.  Methods  The expression of AQP3 in 10 cases with chronic cervical inflammation and 27 cases of cervical carcinoma was determined using real-time PCR, Western blot analysis, and immunofluorescence. The expression of AQP3 protein in 82 cases with cervical carcinoma, 36 cases with Stage-III cervical intraepithelial neoplasia (CINⅢ), and 42 cases with chronic cervical inflammation, was assayed using immunohistochemistry.  Results  AQP3 mRNA and protein were expressed in both the chronic cervical inflammation and cervical carcinoma groups. The AQP3 protein expression was statistically significant (P < 0.05), whereas the AQP3 mRNA expression was not (P>0.05). Moreover, the positive rate of AQP3 was 19.00%, 27.78%, and 41.46% in the chronic cervical inflammation, CIN Ⅲ, and cervical carcinoma groups, respectively, with significant differences among the positive expression (P < 0.05). The expression of AQP3 in cervical carcinoma had no correlation with histological grade, histological type, or lymph node metastasis (P>0.05), however, it had significant association with FIGO stage, diameter of tumor, and invasive depth (P < 0.05).  Conclusion  The expression of AQP3 plays an important role in the carcinogenesis and progression of cervical carcinoma.
