姜黄素对人结肠癌细胞PI3-K/Akt和MEK/ERK信号通路的影响

摘    要:目的:探讨姜黄素对人结肠癌RKO细胞PI3-K/Akt和MEK/ERK通路的影响。方法:MTT法检测细胞活力,Western blot检测p-Akt、Akt、p-ERK,ERK及凋亡相关蛋白Bcl-2、Bax的表达。结果:姜黄素作用人结肠癌RKO细胞,24h和48h的IC50值分别为51.69μg/ml和36.12μg/ml。选用50μg/ml的姜黄素分别作用24h和48h,RKO细胞凋亡百分比为26.79%和42.16%,与对照组相比有显著差异(P<0.05)。进一步检测发现姜黄素(50μg/ml)显著下调了p-Akt 和p-ERK的表达,同时Bcl-2与Bax的比值显著下调。结论:姜黄素可能通过抑制PI3-K/Akt和MEK/ERK信号通路的活化、下调Bcl-2与Bax的比值,从而抑制RKO细胞增殖和诱导细胞凋亡。

Abstract:Objective:To investigate the effects of curcumin on the PI3-K/Akt and MEK/ERK patheway in human colon cancer.Methods:Cell proliferation was measured using MTT assay.The expressions of p-Akt,Akt,p-ERK,ERK and apoptosis-related proteins were detected by Western blot.Results:Curcumin inhibited RKO cell viability and the concertration of inhibited cell viability (IC50) for 24h and 48h were 51.69μg/ml and 36.12μg/ml.After treatment with 50μg/ml curcumin for 24h and 48h,the rates of cell apoptosis were 26.79% and 42.16%,respectively.And after treated 24h with 50μg/ml curcumin,the expressions of p-Akt and p-ERK were significantly down-regulated in RKO cells and the ratio of Bcl-2/Bax expressions was decreased.Conclusion:Curcumin inhibited the activation of the PI3-K/Akt and MEK/ERK signaling pathway,which consequently down-regulated the value of Bcl-2/Bax ratio in RKO cells.
