碘及TSH对甲状腺乳头状癌细胞中miRNA表达的影响

摘    要:目的:体外实验研究人体甲状腺乳头状癌(PTC)细胞中miRNA在碘、TSH影响下的表达变化。方法:体外培养PTC细胞,Western blot法半定量分析p27kip1、AKT、AP-1的表达,RT-PCR法定量分析miRNA-21、miRNA-221、miRNA-222的表达。结果:经不同浓度碘和TSH分别处理PTC细胞,3种miRNA的表达量较对照组均上调;所对应的蛋白p27kip1表达均减少,AP-1及AKT表达较对照组均增加,增加程度与处理因素的浓度未见明显关联。结论:碘、TSH对甲状腺乳头状癌细胞中PI3K/AKT信号通路起到上调作用,从而对甲状腺乳头状癌的发展起到了一定促进作用,且其作用程度与处理浓度无关。

Abstract:Objective:To investigate the expression change of miRNAs under the treatment of iodine and TSH by means of human thyroid papillary carcinoma (PTC) cells culture in vitro.Methods:Cells were cultured for semi-quantitative analysis of p27kip1,AKT,AP-1 expression by Western blot and quantitative analysis of the three miRNAs' expression by RT-PCR.Results:After treated thyroid papillary carcinoma TPC-1 with different concentrations of TSH and iodide separately,miRNA-21,miRNA-221 and miRNA-222 were all up regulated.The miRNAs corresponding protein p27kip1 expression was reduced.Conversely,AP-1 and AKT were increased.All the changes of the expression of the miRNAs and proteins showed no association with concentration of processing factors.Conclusion:Iodine,TSH can increase PI3K/Akt signaling pathway expression in PTC cells,but no significant difference were observed in the expression levels with different concentrations.Both of them may played a catalytic role in the development of papillary thyroid carcinoma.
