摘 要: | [摘要] 目的 探讨白血病患者骨髓血管新生情况及血清血管内皮生长因子(VEGF)水平与临床病情的关系。方法 应用免疫组化染色法(EnVison两步法)检测38例初诊白血病患者治疗前后骨髓活检标本微血管密度(BM-MVD)变化;采用ELISA法测定38例白血病患者治疗前后VEGF的表达,并设对照组进行比较,同时检测骨髓幼稚细胞水平,分析VEGF与白血病类型、病情及预后的关系。结果 初诊白血病患者骨髓MVD[(20.39 ±8.49)/ hpf]明显高于对照组[(8.97±3.49)/hpf](P<0.01);白血病患者的血清VEGF水平明显高于正常对照组(P<0.01)。结论 初诊白血病患者骨髓存在血管新生,骨髓MVD变化与临床病情有关,观察骨髓MVD有助于判断疗效和评估预后;白血病患者VEGF水平明显高于对照组,血清VEGF水平与白血病病情正相关,对白血病的诊断、治疗和预后的判断有一定意义。

Bone Marrow Microvascular Density and Serum Levels of Vascular Endothelial Growth Factor in Patients With Leukemia |

Abstract: | Objective To investigate the correlation between Bone Marrow Angiogenesis ,the level of vascular endothelial growth factors (VEGF) , and disease staging in patients with leukemia. Methods: Bone marrow specimens from 38 patients with leukemia and 40 normal controls were studied.The number of bone marrow microvessels was examined by means of immunohistochemical staining for CD34. Determination of microvesseldensity(MVD)and angiogenes is grading were done in a blinded manner.To detect VEGF in peripheral blood from different stage of leukemia patients. The rate of blast cells and juvenile cells in patients’marrow were assessed simultaneously and analyzed statistically. Bone marrow specimens from 38 patients with leukemia and 40 normal controls were studied.The number of bone marrow microvessels was examined by means of immunohistochemical staining forCD34. Determination of microvesseldensity(MVD)and angiogenesis grading were done in a blinded manner. Results The study showed that bone marrow MVD increased greatly in newly diagnosed leukemia if no treatment was provided. Bone marrow MVD decreased to the same range as the controls at the time of complete remission. Serum levels of VEGF of patients with newly diagnosed acute leukemia increased greatly compared with those of the controls ,but those of the complete remission are significantly elevated compared with the controls and double times higher than the controls. Conclusion Angiogenesis played a role in the bone marrow of newly diagnosed leukemia. Assessment of VEGF for diagnosis and estimation of prognosis in patients with leukemia is of important clinical significance. There was not correlation between serum VEGF levels and bone marrow MVD. |
