缝隙连接增强肿瘤坏死因子-α的肝细胞毒性

摘    要:【目的】 观察缝隙连接(GJ)功能改变对肿瘤坏死因子-α(TNF-α)肝细胞毒性的影响?【方法】 使用3种不同的方法:(1)GJ抑制剂预处理;(2)高低密度接种细胞;(3)小分子RNA转染抑制缝隙连接蛋白32(Cx32)表达等3种不同的方法抑制GJ的功能,观察不同浓度TNF-α作用不同时间后对肝细胞毒性的影响?【结果】 TNF-α的肝细胞毒性呈浓度和时间依赖性;抑制剂预处理后,TNF-α的肝细胞毒性明显降低(P < 0.01);低密度接种细胞(细胞间无GJ形成)时,TNF-α的肝细胞毒性较高密度接种细胞(细胞间有GJ形成)明显降低(P < 0.01);抑制Cx32的表达后,TNF-α的肝细胞毒性明显降低(P < 0.01)? 【结论】 抑制由Cx32组成的GJ的功能后,TNF-α的肝细胞毒性明显降低,Cx32的表达对于TNF-α的肝细胞毒性作用具有重要的作用?

Abstract:【Objective】 To investigate the effects of gap junction on the cytotoxicity of TNF-α in BRL-3A cells. 【Methods】 Three different methods were used to change the function of GJ composed of Cx32 to explore the cytotoxicity of TNF-α on BRL-3A cells: (1) inhibitor pretreatment; (2) cell cultured at different densities; (3) blocking Cx32 expression with siRNA. 【Results】 Cytotoxicity of TNF-α on BRL-3A cells was time and concentration dependent. Cells were pretreated with inhibitors of Cx32, cytotoxicity of TNF-α on BRL-3A cells was decreased significantly (P < 0.01); at low density cell culture (no GJIC formed), cytotoxicity of TNF-α on BRL-3A cells was much lower than that at high density cell culture (GJIC formed) (P < 0.01); blocking Cx32 expression with siRNA also reduced TNF-α cytotoxicity (P < 0.01).
