| 摘 要： | 目的:探讨结肠癌患者外周血中microRNA-630(miR-630)的表达，进一步分析其表达对评估结肠癌进展及预后的价值。方法:使用实时荧光定量PCR（Real time PCR）检测55例结肠癌及21例正常对照者外周血中miR-630的表达，采用统计学方法分析miR-630在结肠癌外周血中的表达及其与肿瘤进展和预后的关系。结果:分析Real time PCR检测结果发现，结肠癌患者外周血中miR-630的表达显著高于正常对照（P<0.05），外周血中miR-630表达与结肠癌的侵袭、转移和分期显著相关（P<0.05），单因素和多因素生存分析发现外周血中miR-630的表达上调与结肠癌预后不良显著相关（P<0.05）。结论:结肠癌患者循环miR-630表达显著上调，与结肠癌侵袭和转移密切相关，并可作为结肠癌的独立预后标记分子。

| Abstract: | Objective:To investigate the expression of microRNA-630(miR-630） in peripheral blood of patients with colon cancer，and to further analyze its value in evaluating the progression and prognosis of colon cancer.Methods:Real-time quantitative PCR was used to detect the expression of miR-630 in peripheral blood of 55 patients with colon cancer and 21 normal controls.The expression of miR-630 in peripheral blood of patients with tumor progression and prognosis was analyzed by appropriate statistical method.Results:The expression of miR-630 in peripheral blood of patients with colon cancer was significantly increased compared with that of normal controls(P<0.05).It was also found to be significantly correlated with tumor invasion，metastasis and TNM stage(P<0.05).Univariate and multivariate survival analysis showed that the up-regulation of miR-630 in peripheral blood was significantly associated with poor prognosis of patients(P<0.05).Conclusion:The expression of circulating microRNA-630 in patients with colon cancer is significantly up-regulated，which is closely related to invasion and metastasis of colon cancer and can be used as an independent prognostic marker for patients with colon cancer. |
