甘氨双唑钠影响宫颈癌SiHa细胞对顺铂的敏感性研究

摘    要:目的:甘氨双唑钠(CMNa)是中国研制的新型放疗增敏剂,对喉癌、鼻咽癌等的放射增敏研究均已取得显著效果,但对化疗药的增敏效果如何,目前仍不肯定。本文以子宫鳞癌SiHa细胞为靶细胞,分析CMNa是否可影响宫颈鳞癌SiHa细胞株对顺铂(PDD)的敏感性。方法:不浓度PDD、CMNa及两药联合作用于SiHa细胞,通过形态学观察、MTT细胞活力测定等方法检测对SiHa细胞敏感性的影响。结果:不同浓度的PDD对宫颈癌细胞的生长均有抑制作用,而不同浓度CMNa对SiHa细胞影响不显著。两药合用可大大增强PDD对SiHa细胞生长的抑制。结论:联合应用CMNa可提高PDD对肿瘤细胞的敏感性,为临床制定宫颈癌化疗方案提供理论依据。

The susceptibilities research of CMNa effecting on cisplatin to cervical carcinoma cell line SiHa
Abstract:OBJECTIVES: CMNa is a novel tumor radiosensitizer,using in laryngeal carcinoma and pharyngeal cancer radiosensitize research, has obtained remarkable effection. The effect of CMNa to the chemotherapy, is still not affirm. This article take the cervical cancer SiHa cell as target cell, analyzes whether CMNa can effects the chemosensitivity of cisplatin on cervical carcinoma SiHa cells. METHODS: SiHa cells were subjected to PDD,CMNa,and both of them in various concentrations for 24hours, and the morphology of the SiHa cells was observed. MTT colorimetry was used to determine the effection of those drugs to SiHa cells. RESULR: The SiHa cells treated with different concentration of cisplatin underwent cell proliferation inhibition. However, various concentration of CMNa are not remarkable effection to the SiHa cells. The combination of CMNa can strengthen the ression of PDD to SiHa cells greatly. ONCLUSION: CMNa can ehance the sensitivity of PDD to SiHa cells,which presumed that CMNa can be used as a chemosensitizer .the research can provide theory basis for formulating cervical cancer chemotherapy plan.
