摘    要:目的:探究lncRNA ZFAS1对人鼻咽癌细胞增殖及凋亡的影响及其调控机制。方法:利用实时荧光定量PCR(RT-qPCR)方法检测鼻咽癌组织和细胞中ZFAS1和miR-135a的表达水平。通过生物信息学分析、双荧光素酶报告分析和RNA免疫沉淀研究ZFAS1与miR-135a表达间的关系。利用LipofectamineTM 2000向鼻咽癌细胞CNE1中单独转染si-ZFAS1、miR-135a mimic,共转染si-ZFAS1和anti-miR-135a及其相关对照,采用CCK-8法、流式细胞术检测转染后细胞的增殖及凋亡能力。结果:鼻咽癌组织和细胞中ZFAS1表达上调,而miR-135a表达下调。miR-135a在鼻咽癌细胞中与ZFAS1直接结合。敲减ZFAS1或过表达miR-135a显著抑制鼻咽癌细胞的增殖,促进细胞的凋亡;而敲减miR-135a表达逆转了ZFAS1沉默介导的鼻咽癌细胞增殖受抑及凋亡受促现象。结论:敲减ZFAS1通过miR-135a抑制鼻咽癌细胞的增殖,诱导凋亡,为鼻咽癌治疗开辟了一条新的途径。

Abstract:Objective:To explore the effect of lncRNA ZFAS1 on proliferation and apoptosis of nasopharyngeal carcinoma cells and its regulatory mechanism.Methods:Real-time fluorescence quantitative PCR(RT-qPCR) was used to detect the expression of ZFAS1 and miR-135a in nasopharyngeal carcinoma tissues and cells.The bioinformatics analysis,dual luciferase report analysis,and RNA immunoprecipitation were used to study the interaction between ZFAS1 and miR-135a.LipofectamineTM 2000 was used to transfect si-ZFAS1,miR-135a mimic separately,co-transfect si-ZFAS1 and anti-miR-135a and their related controls into nasopharyngeal carcinoma cell CNE1.CCK-8 assay and flow cytometry were used to detect cell proliferation and apoptosis after transfection.Results:ZFAS1 expression was up-regulated,while miR-135a expression was down-regulated in nasopharyngeal carcinoma tissues and cells.miR-135a bound directly to ZFAS1 in nasopharyngeal carcinoma cells.Knockdown of ZFAS1 or overexpression of miR-135a significantly inhibited the proliferation,but promoted apoptosis of nasopharyngeal carcinoma cells,while knockdown of miR-135a reversed the suppression of proliferation and acceleration of apoptosis of nasopharyngeal carcinoma cells mediated by ZFAS1 silencing.Conclusion:Knockdown of lncRNA ZFAS1 inhibits proliferation and induces apoptosis of nasopharyngea carcinoma through miR-135a,providing a novel avenue for nasopharyngeal carcinoma treatment.
