摘    要:目的:探究SLC7A11在膀胱癌组织及癌旁组织中的表达差异,并利用基因沉默工具研究SLC7A11对膀胱癌细胞迁移功能的影响。方法:利用免疫组织化学方法检测63例膀胱癌组织和16例膀胱癌旁组织中SLC7A11表达情况;采用统计学方法分析SLC7A11与膀胱癌病理组织参数的关系;RT-qPCR和Western blot方法观察SLC7A11的mRNA和蛋白在膀胱正常上皮细胞和不同类型膀胱癌细胞中的表达;沉默 SLC7A11的表达,划痕、Transwell实验评价SLC7A11对膀胱癌细胞迁移能力的影响。结果:免疫组化结果显示,与癌旁组织比较,SLC7A11高表达于膀胱癌组织中(P<0.05); RT-qPCR和Western blot的实验结果表明SLC7A11异常高表达于膀胱癌细胞T24、BIU-87、5637细胞中;统计分析SLC7A11和膀胱癌的临床病理参数关系,结果揭示SLC7A11的表达与膀胱癌患者的年龄(P=0.612)、性别(P=0.599)、肿瘤大小(P=0.98)、临床TNM分期(P=0.278)无相关性,与膀胱癌的T分期(P=0.038)具有密切关系;斯皮尔曼相关性分析表明SLC7A11与膀胱癌的T分期显著相关(P=0.001);Cox回归单因素分析发现,膀胱癌症患者的死亡风险与膀胱癌的T分期和肿瘤大小相关,与SLC7A11表达和膀胱癌临床TNM分期无相关性,多因素回归分析显示SLC7A11不是膀胱癌生存期的预测标志物。另外,在划痕和迁移实验室中发现,下调SLC7A11的表达,能够明显抑制膀胱癌细胞的迁移能力。结论:SLC7A11高表达于膀胱癌组织和细胞中,能够促进膀胱癌细胞的迁移能力。

Abstract:Objective:To explore the difference of solute carrier family 7 member 11(SLC7A11) expression between bladder cancer tissues and adjacent to bladder carcinoma,and to investigate the effect of SLC7A11 on the migration function of human bladder cancer cells through silencing of SLC7A11 expression.Methods:According to the instruction of immunohistochemical(IHC) staining,the expression of SLC7A11 was examined in the adjacent nonmaligant samples(n=16) and bladder cancer samples(n=62).The association between SLC7A11 expression and clinicopathological parameters of bladder cancer was analyzed.The mRNA and protein levels of SLC7A11 in the normal bladder epithelial cells and bladder cancer cells were detected by RT-qPCR and Western blot,respectively.We silenced SLC7A11 gene in T24 cells and evaluated the migration abilities of bladder cancer cells through wound healing and Transwell migration assay.Results:The results of immunohistochemical showed that SLC7A11 was highly expressed in bladder cancer tissues compared with adjacent to bladder carcinoma(P<0.05).The assay of RT-qPCR and Western blot indicated that SLC7A11 was abnormally high expressed in bladder cancer cells T24,BIU-87,5637 in comparison with bladder normal epithelial cells SVHUC-1.We determined the association between SLC7A11 and the clinicopathological features of bladder cancer,there was closely associations between SLC7A11 expression and T classification(P=0.038).While,the expression of SLC7A11 was not associated with age(P=0.612),gender(P=0.599) or tumor size(P=0.98),clinical stage(P=0.278).Furtherly,Spearman analysis was used to show that the expression of SLC7A11 was dramatically correlated with T classification(P=0.001).Univariate analysis uncovered that the risk of death in bladder cancer patients was associated with T classification and tumor size but was not significantly related to SLC7A11 expression and clinical stage.In addition,multivariate Cox regression analysis determined that SLC7A11 was not a factor for predicting poor survival in bladder cancer.Moreover,infection of LV-SLC7A11 could inhibit the migration power of bladder cancer cells.Conclusion:SLC7A11 high expression in bladder cancer tissues and cells,and could promote the migration ability of the bladder cancer cells.
