| Abstract: | Objective:To investigate the effect of the quinazoline PAK4 small moleculer inhibitor CWS-6 on the proliferation,migration and invasion of pancreatic cancer cells lines.Methods:CCK8 detected the effect of CWS-6 on the proliferation of pancreatic cancer cells at 0 h/24 h/48 h/72 h.The cell cycle progression was observed by Flow cytometry.The effect of CWS-6 on the migration of pancreatic cancer cells was detected by Wound-healing assay.The effect of CWS-6 on the invasion was observed by Transwell assay.Confocal assay observed microfilament of CWS-6.Western blotting assay detected the expression of the activity of PAK4 and also the related proteins with migration and invasion of CWS-6 in pancreatic cancer cells.Results:CWS-6 has no inhibitory effect on the proliferation and cell cycle the of pancreatic cancer cells,but it inhibits the migration and invasion of pancreatic cancer cells at 5 μmol/L.It also suppresses the expression of the activity of PAK4 kinase and the it's downstream proteins relating with cytoskeleton.Conclusion:CWS-6 inhibits cell proliferation,migration and invasion of pancreatic cancer cells by suppressing the activity of PAK4 kinase and the downstream proteins of PAK4 which related to cytoskeleton. |
