Abstract:
The effect of polyglycoside of Tripterygium wilfordii Hook. f. (TWP) on NO production in mouse peritoneal Mф was studied. The results indicated that TWP at 2.0 and 4.0 mg·L-1 (in vitro) and 15 and 60 mg·kg-1 (ig) reduced the NO2- production induced by lipopolysaccharide(LPS) 10 mg·L-1 and interferon-γ(IFN- γ) 150 kU·L-1 in cultured peritoneal Mф obtained from C57BL/6 mice. Concanavalin A(Con A) stimulated Mф produced more NO2- than static Mф and the NO2- level was not affected by LPS, IFN γ or TWP. However, if mice were treated by TWP 15 and 60 mg·kg-1 (ig) 4 d before Con A(ip), the NO2- production in Con A stimulated Mф was lowered. These results suggest that TWP decrease NO production through inhibiting the induction of nitric oxide synthase in Mф, which may partially account for its immunosuppressive actions.
