摘    要:
采用荧光猝灭法研究了叶酸(FA)和去氧胆酸(DH)与牛血清白蛋白(BSA)的相互作用机制,及共存紫杉醇(PA)对FA和DH与BSA结合的影响。实验结果表明,FA和DH与BSA形成复合物而猝灭BSA的内源荧光,荧光猝灭机制符合静态机制;FA和DH与BSA主要靠疏水作用力结合且结合位点都在site Ⅱ;PA对FA和DH的BSA结合位点无竞争,且共存的PA使得FA与BSA结合能力加强,DH与BSA结合能力减弱。

Abstract:
Fluorescence quenching method was used to study the interactions of folic acid and deoxycholic acid(DH) with bovine serum albumin(BSA) with or without coexisting paclitaxel(PA).The results indicated that FA and DH could quench the inner fluorescence of BSA by forming the FA-BSA complex or the DH-BSA complex.Both static quenching and non-radiative energy transfer was confirmed to be the result in the fluorescence quenching.The binding force of FA and DH was mainly hydrophobic force and the binding sites of FA and DH were mainly in the site II.The interaction of FA with BSA was strengthened,and that of DH with BSA was weakened while coexisting with PA.
