摘 要: | 目的探讨两种不同剂量雷洛昔芬(RAL)促排卵对小鼠围着床期子宫内膜胞饮突表达的影响。方法以6~8周龄、连续2个动情周期均正常、未交配过的健康昆明系小白鼠为研究对象,将雌鼠随机分为4组:生理盐水(SS)组、CC组、RAL 180 mg组和RAL 240 mg组,每组12只。在第3个动情周期的动情前期进行灌胃(SS组:灌胃1 mL生理盐水,1次/d,连续2 d;CC 100 mg组:灌胃l mL,CC 18 mg/kg,1 次/d,连续2 d;RAL 180 mg组:灌胃l mL,RAL 33 mg/kg,1 次/d,连续2 d;RAL240 mg组:灌胃lmL,RAL 44 mg/kg,1次/d,连续2 d),2 d后(两次用药后)的下午5:00,分别向每只雌鼠腹腔注射5U HCG,然后分别与正常雄性小鼠1∶1合笼,次日清晨检查阴栓,有阴栓者为受孕Dl。在D4.5采用乙醚吸入麻醉法处死各组雌鼠,取小鼠子宫内膜组织,电镜扫描观察各组子宫内膜胞饮突的发育情况。结果电镜下,RAL 180 mg组、RAL 240 mg组和SS组小鼠子宫内膜胞饮突发育成熟,表达丰富,3 组间两两比较无显著差异,而CC组小鼠子宫内膜胞饮突发育不成熟,数量稀少。胞饮突在RAL 180 mg组、RAL 240 mg组和SS组的半定量表达组间比较无显著差异,均显著高于CC组的表达量。结论两种不同剂量RAL均不影响胞饮突在在围着床期期小鼠子宫内膜上皮的表达,提示两种不同剂量RAL促排卵均不影响小鼠子宫内膜容受性。

Effects of raloxifene at two different doses for ovulation induction on endometrialpinopodes in mice during the implantation window |

Abstract: | Objective To compare the expression of pinopodes, the marker of endometrial receptivity, during the implantationwindow in Kunming mice stimulated with two different doses of raloxifene (RAL). Methods Forty-eight 8-week-old femaleKunming mice were randomly divided into 4 groups (n=12), namely saline group, clomiphene citrate (CC, 18 mg/kg) group,RAL (33 mg/kg) group and RAL (44 mg/kg group). In each group, the mice received intragastric administration of 1 mL ofnormal saline containing CC or RAL at the specified doses or saline only as indicated for ovulation induction, once daily for 2days. The mice received then injection with 5 IU human chorionic gonadotropin (HCG) and mated and on day 4.5 of gestation,the pregnant mice were sacrificed for examination of the uterus with scanning electron microscopy. Results Abundant andwell developed pinopodes were observed in the endometrium of the mice in the 2 RAL groups and in the saline control group.The mice in CC group showed obviously reduced endometrial pinopodes with poor development. Conclusions RAL at twodifferent doses does not obviously affect the expression of pinopodes in the uterine epithelium of mice, suggesting the safetyof RAL at these two doses for ovulation induction without causing adverse effects on endometrial receptivity. |
