丝裂霉素C对阻塞性泪道MEK/ERK信号通路的影响

摘    要:目的 探讨丝裂霉素C(mitomycinC,MMC)对阻塞性泪道MEK/ERK信号转导通路及其相关蛋白表达水平的影响,旨在为临床阻塞性泪道疾病的诊治提供实验依据。方法 30只健康家兔,分为正常对照组(NC组)、模型组、Nd-YAG激光组(Y组)、MMC组及Nd-YAG激光+MMC组(Y+M组),每组各6只。通过球囊导管摩擦泪道建立阻塞性泪道模型,Y组、MMC组、Y+M组家兔分别给予Nd-YAG激光、MMC、Nd-YAG激光联合MMC治疗,治疗3个月后处死,光镜观察泪道上皮组织形态学变化,免疫组织化学法测定各组PCNA、ERK1、ERK2、MMP-3、MMP-9阳性细胞面积。分离阻塞性泪道细胞,随机分为空白组、阴性对照组(加入DMSO)及阳性荧光组(加入MEK抑制剂PD98059),采用WesternBlot检测各组细胞ERK1、ERK2表达情况。结果 模型组PCNA、ERK1、ERK2、MMP-3、MMP-9阳性细胞面积均大于NC组,差异均有统计学意义(均为P<0.05);Y组、MMC组、Y+M组PCNA、ERK1、ERK2、MMP-3、MMP-9阳性细胞面积均显著小于模型组,其中Y+M组小于Y组及MMC组,差异均有统计学意义(均为P<0.05)。空白组、阴性对照组、阳性荧光组ERK1的相对表达量分别为152.32±7.45、149.36±7.96、64.32±5.23,ERK2的相对表达量分别为154.98±6.78、151.22±8.23、65.45±3.45,阳性荧光组中ERK1、ERK2蛋白表达水平显著低于空白组及阴性对照组,差异均有统计学意义(均为P<0.05)。结论 Nd-YAG激光与MMC联合应用可增加激光泪道成形术治疗效果,其作用机制可能与MMC阻断MEK/ERK信号转导通路有关。

Abstract:Objective To investigate the effects of mitomycin C ( MMC ) on MEK/ERK signal transduction pathway and levels of related protein expression in obstructive lacrimal, and provide an experimental basis for clinical diagnosis and treatment of obstructive lacrimal disease. Methods Thirty healthy rabbits were divided into normal control group ( NC group ) , model group , Nd-YAG laser group ( Y group ) , MMC group and Nd-YAG laser + MMC group (Y +M group) ,6 cases in each group. The rabbit models of obstructive lacrimal were established by rubbing a balloon catheter. the rabbits in Y group , MMC group and Y + M group received the Nd-YAG laser , MMC .Nd-YAG laser + MMC .then sacrificed after 3 months of treatment , light microscopy was used to observe the lacrimal morphological changes. PCNA. ERKI , ERK2 , MMP-3 , MMP-9 positive cell area were measured in each group by immunohistochemistry. Separation obstructive lacrimal cells were randomly divided into blank group , negative control group ( adding DMSO) and positive fluorescent group (joirung MEK inhibitor PD98059) .Western Blot was used to detect the expression of ERKI .ERK2 expression. Results PCNA.ERKI , ERK2 .MMP-3 , MMP-9 positive cell area in model group were larger than those in NC group ( all P < 0. 05 ) . PCNA , ERKI .ERK2 , MMP-3 , MMP-9 positive cell area in Y group , MMC group ,Y + M group were significantly less than those in model group ,while Y + M group was less than Y group and MMC group ( all P < 0. 05 ) . The relative expression of ERKI in blank group , negative control group and positive fluorescent group were 152. 32 +7. 45 . 149. 36 +7. 96 .64. 32 + 5. 23 ,respectively , ERK2 were 154. 98 + 6. 78 , 151. 22 + 8. 23 ,65. 45 +3. 45 .the relative expression of ERKI and ERK2 in positive fluorescent group were obviously lower than those in blank group and negative control group ( all P < 0. 05 ) . Conclusion Nd-YAG laser combined with MMC increase the efficacy of lacrimal laser angioplasty, and its mechanism may be related to MMC blocking MEK / ERK signal transduction pathway.
