摘    要:摘 要:目的:分离培养鼠骨髓间充质干细胞(BMSCs)通过脑室注射移植给缺血缺氧性脑损伤(HIBD)大鼠,观察其对大鼠影响。方法:治疗组予脑室注射BMSCs,对照组予脑室注射等量生理盐水,免疫组织化学法进行检测。结果:在受损半球,治疗组各个时间点 VEGF表达强度及微小血管密度显著高于对照组。在移植后第1d,对照组及治疗组VEGF表达强度无差异。第3、7d治疗组VEGF表达强度及新生小血管均多于对照组,至2周两组再次恢复到相同水平。在治疗组大鼠脑组织切片中可以看到Brdu阳性细胞。结论:BMSCs通过脑室内注射的进入脑组织成活并定植,可增强VEGF表达。外源性的细胞移植能促进损伤脑组织血管再生,从而减轻脑组织缺氧缺血损伤的程度。

Abstract:ABSTRACT: To explore the angiogenesis effect of BMSCs from human umibilical cord blood after transplantation into rat brain. Method:The BMSCs were transplanted into the rats brain in therapy group. control group with and HIBD group with narmal saline. Result:In immunohistochemical detection, the expressing of VEGF in HIBD and therapy group were higher than in control group in day 1, 3, 7, 14. The expression strength of VEGF in HIBD and therapy group were not significantly different, and which was higher in therapy groups than in HIBD group in day 3 and 7, it descended in both groups in day 14. Brdu positive cells could be detected in the rat brains afrer 14d of transplantation. Accordingly we got a conclusion . The BMSCs marked with Brdu were transplanted into the brain of HIBD rats by stereotaxic apparatus can permanent plante in rat brain. Conclusion:The VEGF expression in animal brain would be up regulate after Hypoxic-ischemic damage. Transplantation of BMSCs showed the ability make play a positive role on vascular regeneration.
