合成甘氨酸锌复合佐剂对鸡血清蛋白抗原诱导小鼠免疫应答的影响

摘    要:目的 探讨合成甘氨酸锌复合佐剂对鸡血清蛋白抗原诱导小鼠体液免疫和细胞免疫的影响。方法 将昆明ICR雌性小鼠随机分为 4组:复合佐剂组(甘氨酸锌20μg+铝佐剂80μg+100μg抗原)、空白对照组(生理盐水100μl+100μg抗原)、甘氨酸锌对照组(甘氨酸锌20μg+100μg抗原)和阳性对照组(铝佐剂80μg+100μg抗原),每组6只。各组均经腹部皮下多点注射,100μl/只,共免疫1次。免疫4、8、12、16周后,采用ELISA 法检测小鼠血清IgG滴度、细胞因子IL-10和IL-12,并进行安全性试验。结果 免疫后第8周时,复合佐剂组的抗体水平与阳性对照组和甘氨酸锌对照组有统计学差异(t=9.189,t=16.838,P 均< 0. 05),第12和16周时,复合佐剂组的抗体水平与阳性组的差异无统计学意义(t=1.794,t=0.543,P>0.05);8周时复合佐剂组的IL-10和IL-12水平与阳性对照组和甘氨酸锌对照组有统计学差异(P均<0. 05);病理切片显示,复合佐剂组心、肝、脾、肺、肾等器官未出现毒性病理反应。结论 合成甘氨酸锌与铝佐剂的复合佐剂能增强鸡血清蛋白诱导小鼠的体液免疫和细胞免疫应答反应。

Abstract:To investigate the effect of zinc glycine complex adjuvant on humoral and cellular immunity induced by chicken serum protein antigen in mice. Methods Kunming ICR female mice were randomly divided into 4 groups: compound adjuvant groups ( 20 μg zinc glycine + 80 ug aluminum adjuvant + 100 μg antigen) ,normal saline control group ( 100 μl normal saline + 100 μg antigen) ,zinc glycine control group ( 20 μg zinc glycine + 100 μg antigen) and positive control group( 80 μg aluminum adjuvant + 100 μg antigen) . Each group received subcutaneous multi - point abdominal injection,100 μl per unit,and were immunized once in total. After immunization at 4,8,12 and 16 weeks,serum IgG titer,cytokines IL - 10 and IL - 12 in mice were detected by ELISA,and safety tests were performed. Results At 8 weeks after immunization,the antibody level of the compound adjuvant group was different from that of the positive control group and the zinc glycine control group ( t = 9. 189,t = 16. 838,both P < 0. 05) . At 12 and 16 weeks,the antibody level of the compound adjuvant group was not different from that of the positive group ( t = 1. 794,t = 0. 543,P > 0. 05) . At 8 weeks,the levels of IL - 10 and IL - 12 in the compound adjuvant group were different from those in the positive control group and the zinc glycine control group ( both P <0. 05) . Pathological sections showed that no toxic pathological reactions were observed in the heart,liver,spleen,lung,kidney and other organs of the compound adjuvant group. Conclusion The combination of zinc glycine and aluminum adjuvant can enhance the humoral and cellular immune responses induced by chicken serum protein in mice.
