摘    要:目的:研究AQP4在人乳腺癌细胞中的表达,并进一步探讨AQP4对肿瘤细胞增殖的影响及机制。方法:通过 siRNA干扰技术下调乳腺癌细胞T47D 和 MCF-7 细胞系中水通道蛋白4表达,检测这种下调对乳腺癌细胞增殖的影响并初步探讨其作用机制。结果:AQP4表达下降抑制乳腺癌细胞的增殖,可能通过ERK通路/E-钙黏蛋白实现的。结论:AQP4表达水平与乳腺癌细胞增殖正相关,相应的肿瘤抑制剂可以起到控制肿瘤生长作用。

Abstract:Objective:To study the expression of AQP4 in human breast cancer cells,and further explore the effect and mechanism of AQP4 on the proliferation of tumor cells.Methods:Down regulation of aquaporin 4 expression in breast cancer cell lines T47D and MCF-7 was performed by siRNA interference technology.The effect of this downregulation on the proliferation of breast cancer cells was detected and its mechanism was explored.Results:The decrease of AQP4 expression inhibited the proliferation of breast cancer cells.It may be realized through the E-calcium/ERK pathway.Conclusion:AQP4 expression level is positively related to the proliferation of breast cancer cells.The corresponding tumor inhibitors can play a role in controlling tumor growth.
