Abstract: | Objective To explore the passage adaptability of yellow fever virus (YFV)17D attenuated strain in human diploid cell (2BS strain). Methods The proliferation of YFV17D in 2BS cells was observed under various conditions and continuous passage. Virus titer was measured by plaque detection. Results YFV17D adapted to the growth in 2BS cells after five continuous passage culture. YFV at multiplicity of infection of 0.005-0.010 reached a highest titer after culture at 35-37 ℃ in determined, pH7.4 medium . Conclusion The adaptability of YFV17D on human diploid cell 2BS is determined, which lays a foundation of preparation of yellow fever vaccine used with human diploid cells. |
