西红柿汁体外抑制血小板聚集功能及其机制探讨

摘    要:目的:观察西红柿汁在体外对血小板聚集、血小板活化标志物的表达及对血小板纤维蛋白原结合量的影响,初步探讨西红柿汁对血小板聚集功能的影响机制。方法:将不同浓度的西红柿汁与正常人富血小板血浆(PRP)孵育,加入5-腺苷二磷酸二钠盐(ADP)和胶原,观察西红柿汁对血小板聚集的影响。用ADP及胶原激活血小板,流式细胞仪(FCM)分析西红柿汁对活化血小板纤维蛋白原受体(Fib-R)和P-选择素(CD62P)的表达水平及血小板纤维蛋白原结合量的影响。结果:西红柿汁能够明显抑制由ADP、胶原诱导的血小板聚集,且随着西红柿汁浓度的增加,对血小板聚集的抑制率增高。西红柿汁不能抑制ADP、胶原诱导的血小板Fib-R、CD62P表达,但能显著抑制由ADP、胶原诱导的血小板与纤维蛋白原结合量。结论:西红柿汁在体外能够显著抑制血小板聚集,其抗血小板聚集的作用机制与阻碍纤维蛋白原与血小板Fib-R结合有关。西红柿汁具有潜在的抗血小板治疗前景。

Abstract:Objective To observe the effect of tomato juice on the human platelet aggregation in vitro, the expression of the platelet activation marker and the effect of the binding of fibrinogen and platelet fibrinogen receptor(Fib-R), and to elucidate the mechanism of the tomato juice effects on the human platelet aggregation. Methods The tomato juice was mixed with the platelet-rich plasma (PRP) in different proportion, and the maximal ratio of platelet aggregation induced by 5-adenosine liphosphate-disodium salt (ADP) and collagen was detected on the aggregometer. After the platelet was activated by ADP and collagen, the expression levels of the platelet activation markers including Fib-R and CD62P (P-selectin), and the combination amount of the platelet Fib-R and the fibrinogen were examined by flow cytometry. Results The tomato juice significantly inhibited the platelet aggregation induced by ADP and collagen, and the inhibition intensity to the platelet aggregation elevated with the increase of the tomato juice concentration. Although the tomato juice could not inhibit the expressions of Fib-R and CD62P induced by ADP and collagen on the activated platelet membrane surface, it was able to decrease significantly the combination of ADP- and collagen-induced Fib-R and the fibrinogen. Conclusions Tomato juice can inhibit significantly the platelet aggregation in vitro. As an antagonist of the platelet fibrinogen receptor, the mechanism of tomato juice inhibiting the platelet aggregation is possibly related to the interruption of the interaction between the platelet Fib-R and the fibrinogen. Tomato juice is promising in the anti-platelet therapy.
