骨碎补总黄酮对大鼠牙髓干细胞增殖和细胞周期的影响

摘    要:摘要 目的 探讨骨碎补总黄酮对大鼠牙髓干细胞(DPSCs)增殖和细胞周期的影响。方法采用组织块消化法获得大鼠牙髓细胞,克隆化分离培养大鼠DPSCs并进行鉴定。以含0.01,0.05和0.10 g·L-1骨碎补总黄酮的培养基分别培养大鼠DPSCs,采用噻唑蓝(MTT)比色法检测各组细胞的增殖情况,流式细胞术检测各组细胞周期的变化。结果经单克隆化分离培养得到的DPSCs细胞CD44、CD29和Stro 1均呈阳性表达。骨碎补总黄酮组DPSCs增殖速度较对照组加快(P<0.05),且随骨碎补总黄酮浓度的增加而升高。流式细胞术分析表明S期细胞比例明显多于对照组(P<0.05),G0/G1期细胞明显减少(P<0.05),且表现出剂量依赖性。 结论骨碎补总黄酮对大鼠DPSCs增殖具有促进作用,这一作用可能是通过促进DPSCs从G0/G1进入S期来实现。

Abstract:Objective To explore the effects of total flavonoids of drynaria on the proliferation and cell cycle of rat dental pulp stem cells.Methods The rat dental pulp stem cells(DPSCs) were obtained with a tissue digesting method,which were cloning isolated and cultured for further identification.The dental pulp stem cells were cultured in media with different concentrations of drynaria total flavonoids(0.01 g·L-1,0.05 g·L-1 and 0.10 g·L-1).Then the proliferation was detected by MTT,and the changes of cell cycle in each group were analyzed by flow cytometry.Results CD44,CD29 and Stro-1 were positive in DPSCs.The proliferation rate of DPSCs treated with the total flavonoids of drynaria was accelerated in comparison to the control group(P<0.05).The proportion of cells in phase S was conspicutously more than that in the control group(P<0.05),and cells at G0/G1 phase were significantly decreased(P<0.05),which were also showed in a dose-dependent manner.Conclusion The total flavonoids of drynaria could promote the proliferation of DPSCs in rats,and the effect of which might rely on expediting DPSCs from phase G0/G1 into phase S.
