摘    要:
目的建立分离提取高纯度卡那霉素B的最佳工艺条件。方法以基因工程黑暗链霉菌spu46发酵产物为原料,通过对树脂吸附性能的研究,筛选吸附容量最大的弱酸性树脂;考察上样液的pH、浓度、流速等因素对吸附效果的影响;考察不同洗脱条件对分离提纯效果的影响。结果弱酸性阳离子树脂CD-180在pH 7.5、流速0.85 mL.m in-1的条件下,吸附量达到最高。0.15 mol.L-1的氨水在1.00 mL.m in-1,的流速下,解析率稳定,可达到85%以上。结论弱酸性阳离子树脂CD-180对卡那霉素B的分离纯化效果最佳。

Abstract:
Objective To determine the best extraction process of kanamycin B. Methods Based on the fermentation broth of Streptomyces tenebrarius spu46 producing kanamycin B, static adsorption and dynamic adsorption were studied to choose resin with the best adsorption capability. Effect of pH value, concentration and flow rate on performance of absorption was investigated using the chosen resin. Effect of different desorption parameter on extraction was also researched. Results Weakly acidic cationic resin had the best performance of absorption with pH7.5 and flow rate of 0.85 mL•min-1. Desorption rate could reach 85% when concentration of ammonia water was 0.15 mol•L-1, with flow rate of 1.00 mL•min-1. Conclusion Weakly acidic cationic resin CD180 has the best capability on the extraction of kanamycin B.
