长链非编码RNA-ROR与Nrf2在亚砷酸钠慢性染毒人支气管上皮细胞中的表达与关系

摘    要:目的 以亚砷酸钠慢性染毒人支气管上皮(HBE)细胞,观察长链非编码RNA-ROR(lncRNA-ROR)与核转录因子-E2类相关因子2(Nrf2)在砷慢性染毒细胞中的变化规律,并分析二者的相关性。方法 以2.5 μmol/L 亚砷酸钠染毒HBE细胞共40代,分别用实时荧光定量PCR法和蛋白免疫印迹实验检测染毒0代(P0)、10代(P10)、20代(P20)和40代(P40)细胞中lncRNA-ROR的表达水平和Nrf2的蛋白表达水平,并用Pearson相关检验分析lncRNA-ROR和Nrf2的相关性。结果 在亚砷酸钠慢性染毒HBE细胞的过程中,lncRNA-ROR水平逐渐增加,Nrf2蛋白表达水平逐渐降低。lncRNA-ROR在P10细胞中的表达为P0细胞的1.17倍(P>0.05),在P20和P40细胞中的表达分别为P0细胞的1.61倍和2.18倍,差异均具有统计学意义(P<0.05)。Nrf2在P10、P20和P40细胞中的表达分别为P0细胞的0.85倍、0.68倍和0.56倍,差异均具有统计学意义(P<0.05)。lncRNA-ROR的表达水平与Nrf2的蛋白表达水平呈负相关,相关系数r=-0.966。结论 亚砷酸钠慢性染毒能持续抑制Nrf2的蛋白表达水平,进而激活lncRNA-ROR的表达,提示lncRNA-ROR与Nrf2可能共同参与了砷致肿瘤的发生发展。

Abstract:Objective To explore the change rules of non-coding RNA-ROR and the nuclear factor-E2 related factor2 (Nrf2) in the bronchial epithelial cell induced by sodium arsenite (HBE),between which the correlation was analyzed.Methods A total of 40 generation of HBE cells were continuously exposed to 2.5 μmol/L sodium arsenite.RT-PCR and western blot were performed to detect the expression levels of lncRNA-ROR and Nrf2 in cells P0,P10,P20 and P40.Then Person correlation analysis was used to analyze the correlation between lncRNA-ROR and Nrf2.Results The level of lncRNA-ROR gradually increased while the expression of Nrf2 in cells gradually decreased with the exposure of sodium arsenite.Specifically,the level of lncRNA-ROR in P10 cells was 1.17 times than that in cells P0(P>0.05).In cells P20 and P40,the levels of lncRNA-ROR were 1.61 and 2.18 times than that in cells P0 (P<0.05).Moreover,the expression levels of Nrf2 in cells P10,P20 and P40 were 0.85,0.68 and 0.56 times than that in P0 cells (P<0.05).The lncRNA-ROR level was negatively correlated with the expression level of Nrf2,and the correlation coefficient was -0.966.Conclusion Chronic sodium arsenite exposure may suppress the expression level of Nrf2 and activate the expression of lncRNA-ROR,thus leading to the tumor occurrence and progression.
