| 摘 要： | 目的 建立超高效液相色谱-串联质谱同位素内标法测定小龙虾虾肉及虾黄中孔雀石绿及隐色孔雀石绿的残留量。方法 样品经匀浆后加入内标和乙腈提取，提取液经中性氧化铝净化。净化液浓缩后，采用电喷雾（ESI+）多反应监测（MRM）模式进行检测。结果 孔雀石绿及隐色孔雀石绿在0.10～2.00 ng/ml浓度范围内线性关系均良好，相关系数均大于0.999，方法检出限在0.01～0.04 μg/kg之间，平均回收率在92.1 %～104.5 %之间，相对标准偏差（RSD）在2.2 %～7.9 %之间。结论 该方法简便灵敏重现性好，可适用于小龙虾虾肉及虾黄中的孔雀石绿和隐色孔雀石绿的测定。

| Abstract: | Objective To establish a method for determination of malachite green (MG) and its metabolite leucomalachite green (LMG) in crayfish by ultra-high performance liquid chromatography tandem mass spectrometry (LC-MS/MS). Methods After homogenization, the analytes in sample was extracted with acetonitrile. Then the extracting solution was purified with neutral alumina column, and finally separated and detected by LC-MS/MS with electrospray ionization(ESI+) mode and multiple reaction monitoring (MRM) mode. Results MG and LMG showed good correlation in the range of 0.10-2.00 ng/ml, with the correlation coefficient of 0.999. The average recoveries for MG and LMG in crayfish were 92.1%-104.5% with relative standard deviation (RSD) of 2.2%-7.9%. The detection limit of MG and LMG were 0.01μg/kg-0.04μg/kg. Conclusion The method is simple, sensitive and reproducible, and it is suitable for the determination of MG and LMG in crayfish. |
