中文摘要:目的 在SD大鼠头端延髓腹外侧区(rostral ventrolateral medulla,RVLM)内给予salusin-β,探究其对大鼠心率(heart rate,HR)和血压(blood pressure,BP)的影响。方法 雄性SD大鼠32只,随机分为5组,单侧RVLM微量给予谷氨酸(L-glutamic acid,Glu)、人工脑脊液(artificial cerebrospinal fluid,aCSF)或不同剂量的salusin-β(0.04、0.4、4pmol),监测salusin-β对SD大鼠HR和BP的影响。结果 大鼠RVLM微量给予aCSF不影响麻醉大鼠的BP 1±1mmHg,给予Glu血压升高显著10±1mmHg(P<0.05)。RVLM微量给予salusin-β(0.04、0.4、4pmol)均产生升高血压的作用,且呈剂量依赖性(0.04pmol:6±1mmHg,P>0.05;0.4pmol:12±1mmHg,P<0.05;4pmol:29±3mmHg,P<0.05),且4pmol组与Glu组比较,差异有统计学意义(P<0.05)。但salusin-β对心率无明显影响。结论 salusin-β在SD大鼠RVLM产生剂量依赖性升高BP的作用,但对HR无明显影响。 |

Abstract:Objective To investigate the effects of salusin-β on blood pressure and heart rate within the rostral ventrolateral medulla in Sprague-Dawley rats. Methods Thirty-two adult male Spargue-Dawley rats were randomly divided into five groups. The dose-dependant effects of salusin-β(0.04, 0.4 and 4pmol, respectively) on blood pressure and heart rate in the RVLM were determined by unilateral microinjection of salusin-β, L-glutamic acid or artificial cerebrospinal fluid(aCSF) into the RVLM. Results Unilateral microinjection of aCSF into the RVLM did not affect blood pressure 1±1mmHg in anesthetized rats, injection Glu significantly elevated blood pressure 10±1mmHg(P<0.05). Unilateral microinjection of salusin-β into the RVLM produced a dose-dependant increase in blood pressure(BP)(0.04pmol:6±3mmHg, P>0.05; 0.4pmol:12±3mmHg, P<0.05; 4pmol:29±8mmHg, P<0.05), and with little effect on heart rate(HR). Conclusion Salusin-β in RVLM can cause dose-dependant elevation of BP, but no significant effect on HR. |
