睡眠剥夺及睡眠恢复后大鼠中缝核群星形胶质细胞的反应及其与神经元的关系

摘    要:目的探索睡眠剥夺及睡眠恢复后大鼠脑干中缝核群星形胶质细胞的反应及其与神经元的关系。方法采用小平台水环境法建立大鼠睡眠剥夺模型,20只大鼠分为睡眠剥夺12h组,睡眠剥夺12h恢复睡眠3h组及大平台对照组和正常单独饲养组,每组5只,用免疫组化的方法测量glial fibrillary acidic protein(GFAP)和Fosprotein在脑干中缝核群的表达。结果睡眠剥夺后GFAP-like immunoreaction(LI)星形胶质细胞和FosLI神经元在中缝核群各核有明显表达,睡眠恢复后表达明显减少,两者的变化趋势基本一致。结论睡眠剥夺影响GFAPLI星形胶质细胞和FosLI神经元的表达,提示星形胶质细胞和神经元可能共同参与睡眠调节。

Abstract:Objective To investigate the response of the raphe nucleus astrocyte to sleep deprivation and sleep recovery,as well as its relationship with neurons. Methods 20 Sprague-Dawley rats were divided to four group. Two group were sleep deprived by housing them on the small platforms over water for 12 h. After that one group can sleep 3 h.Control were housed in the normal cageor on the large platform over water. The effects of sleep deprivation and sleep recovery on the expression of glial fibrillary acidic protein (GFAP) and Fos protein by immunocytochemistry were investigated. Result Sleep deprivation resulted in increasing expression of GFAP-like immunoreaction (-LI) astrocytes and Fos-LI neurons in the raphe nucleus. After sleep, the expression of GFAP-like immunoreaction (-LI) astrocytes and Fos-LI neurons decreased significantly. The change is similar to the expression of c-fos protein. Conclusion Sleep deprivation may affect GFAP-like immunoreaction (-LI) astrocytes and Fos-LI neurons expression, astrocytes and neurons involved in the regulation of sleep together.
