丁苯酞注射液对慢性脑低灌注状态大鼠前脑线粒体超微结构及ATP酶活性的影响

摘    要:目的:观察丁苯酞注射液对慢性脑低灌注状态大鼠前脑线粒体超微结构及ATP酶活性的影响。方法:雄性SD大鼠30只,随机分为假手术组,生理盐水组,丁苯酞组。术后第4周,生理盐水组腹腔注射生理盐水;丁苯酞组腹腔注射丁苯酞注射液。治疗4周后,观察电镜下线粒体超微结构改变;提取线粒体,测定线粒体成分的ATP酶活性。结果:电镜观察表明丁苯酞组线粒体超微结构的破坏明显比生理盐水组轻。丁苯酞组的Na-K-ATP酶活力明显高于生理盐水组(P<0.01),丁苯酞注射液可明显增加线粒体ATP酶活性。结论:丁苯酞注射液对慢性脑低灌注状态下线粒体结构和功能具有一定保护作用。

Abstract:Objective:To observe effects of dl-n-butylphthalide(dl-NBP)on mitochondria ultrastructure and the activity of mitochondria ATPase in Rats with Chronic Cerebral Hypoperfusion.Methods:30 Sprague-Dawlay male rats,were randomly divided into sham group,saline group and dl-NBP group.After 4 weeks following the operation,dl-NBP group were injected dl-NBP(10mg/kg/d) through abdomen cavity for 4 weeks;Saline group were injected isodose saline.4 weeks later,the changes of mitochondria ultrastructure was observed by electron microscopy and then the mitochondria was collected and the activity of mitochondria ATPase were determined.Results:More mitochondria injury was observed in Saline group than that in dl-NBP group.The activity of ATPase in the Saline group(2.67 ± 0.41μmolPi/mgprot/hour) was significantly lower than that in dl-NBP group(3.41 ± 0.45μmolPi/mgprot/hour)(P 〈 0.01).Conclusion:The mitochondria structure and function in Rats with Chronic Cerebral Hypoperfusion can be protected by dl-NBP.
