盐酸多奈哌齐对拟VaD大鼠海马CA1区NR1及NR2B免疫组织化学表达的影响

摘    要:目的探讨盐酸多奈哌齐对拟血管性痴呆大鼠海马神经元N-甲基-D-天门冬氨酸(NMDA)受体亚单位R1(NMDAR1,NR1)和R2B(NMDAR2B,NR2B)的免疫组织化学表达的影响。方法采用双侧颈总动脉反复夹闭、再通,并腹腔注射硝普钠法制备模型,用盐酸多奈哌齐溶液灌胃,Y-型迷宫试验观察其行为学改变,用免疫组织化学技术观测大鼠海马神经元NR1、NR2B的表达变化。结果盐酸多奈哌齐组大鼠海马CA1区NR1表达明显降低,与模型组比较有显著性差异(均P<0.01),与假手术组相比差异无显著性;NR2B表达较模型组明显增高(P<0.01),与假手术组比较无显著性差异。结论盐酸多奈哌齐有可能通过降低NR1的表达,提高海马NR2B的表达,从而改善拟血管性痴呆大鼠的学习、记忆成绩。

Abstract:Objective To probe the effect of donepezil on N-methy1-D-aspartate receptor subunit proteins NR1 and NR2B in neuron of hippocampus in mice with vascular dementia.Methods The mice were subjected for reperfusion repeatedly on bi-lateral common carotid arteries and injected sodium nitroprusside into the abdominal cavity to establish the imitative vascular dementia rat models.Donepezil was given to the mice in medication group.The behavioral abnormalities were observed by Y-type maze test,and the expressions of N-methy1-D-aspartate receptor subunit proteins NR1 and NR2B in neuron of hippocampus were detected by immunohistochemistry technique.Results Compared with model group,the expression of N-methyl-D-aspartate receptor subunit proteins NR1 was decreased significantly(P<0.01),no significant was found between Donepezil group and sham-operated group,but the expression of N-methy1-D-aspartate receptor subunit proteins NR2B was increased evidently than model group (P<0.01).No difference was found between Donepezil group and sham-operated group.Conclusions Decreased expression of N-methy1-D-aspartate receptor subunit proteins NR1 and increased expression of NR2B may be involved in the improvement of learning and memory in mice with vascular dementia.
