经典的小鼠大脑中动脉闭塞再灌注模型的建立和评价

摘    要:目的:建立经典的小鼠大脑中动脉闭塞再灌注模型。方法:以头端涂硅胶的尼龙线自左颈外动脉向颈内动脉插入至大脑中动脉起始部,阻断血流2h后拔出线栓再灌注。通过神经功能评分和氯化三苯基四氮唑(tetrazoliumchloride,TTC)染色对模型进行评价。结果:小鼠缺血2h后,出现自主活动时向右旋转等功能障碍表现,TTC染色显示出梗死范围。再灌后22hTTC改变加重,神经病学评分同前。结论:该模型可以控制缺血和再灌注时间,是研究脑栓塞病理生理等的经典动物模型。

Abstract:Aim: To establish a classical model with middle cerebral artery occlusion reperfusion (MCAO/ R) in mice. Methods: The nylon suture with silica gel covered at an end was sent from the left external carotid artery into internal carotid artery till arriving at the initiation of middle cerebral artery to block the blood stream, then drawing the suture from ICA 2 hours after occlusion to accomplish reperfusion. The model was evaluated by neurology score and tetrazolium chloride (TTC) stain. Results: The mice revolved to the right side on autonomatic activities 3 hours after middle cerebral artery occlusion. TTC stain showed the infarction region clearly. 22 hours reperfusion after occlusion of blood stream for 2 hours aggravated the TTC changes, while the neurology score remained the same. Conclusion :This model is stable with accurate timing control of occlusion and reperfusion. It is the classical one for researching of pathophysiology in cerebral embolism.
