大鼠冷冻性脑损伤动物模型的建立及评价

摘    要:目的建立稳定的大鼠冷冻性脑损伤动物模型并进行评价。方法以自制冷冻杯,用液氮进行大鼠硬脑膜外冷冻,监测伤后的颅内压变化,测定伤后不同时期的行走、平衡等行为功能,并行神经组织病理检查。结果大鼠冷冻伤后出现颅内压升高,伤后4h达到高峰(14.56±2.31)mmHg;伤后的行走和平衡功能明显障碍;冷冻性脑损伤后神经细胞周围出现水肿,同时伴有脑皮质冻伤灶神经元变性、坏死,神经细胞和神经胶质细胞的空泡样变。结论大鼠的冷冻性脑损伤模型是一种稳定可靠的脑损伤动物模型。

Abstract:Objective To establish and evaluate of cryogenic brain injury rat model. Methods Rat dural external was froze by liquid nitrogen. The changes of intracranial pressure was monitored, the behavior function such as walking, balance function at various period after traumatic brain injury was measured and the pathologic study of nerve tissue was detected. Results Intracranial pressure of cryogenic brain injury group was elevated significantly after cryogenic injury, the maximum value of (14.56 ±2.31) mmHg appeared 4 h after injury. The walking and balance function was evidently hindered. The nerve pathologic study showed that the edema was found around the nerve cells after cryogenic brain injury. Simultaneously neurons degeneration and necrosis presented in brain cortex congelation injury foci. The nerve cells and glial cells were like vacuoles. Condusion The cryogenic brain injury rat model is a stable and reliable brain injury animal model.
