无抽搐电休克对抑郁大鼠学习记忆功能的影响及其突触可塑性机制

摘    要:目的探讨无抽搐电休克对抑郁大鼠学习记忆功能的影响及其突触可塑性机制。方法SD大鼠50只随机分为无抽搐电休克(MECT)组、电休克(ECT)组、丙泊酚组、抑郁模型组(简称抑郁组)、正常对照组(简称对照组)各10只,前4组采用孤养结合慢性不可预见轻度应激制备抑郁模型。建模成功后,MECT组和ECT组每日1次分别腹腔注射丙泊酚和生理盐水后给予电刺激,丙泊酚组腹腔注射丙泊酚但不给予电刺激,连续7d。采用openfield和Morris水迷宫分别评定行为学和学习记忆功能的改变,免疫组织化学法和逆转录聚合酶链反应检测海马突触素(SYP)及其mRNA的表达。结果治疗后MECT组和ECT组的openfield评分均分别明显高于丙泊酚组和抑郁组,差异有统计学意义(P0.05);ECT组Morris水迷宫测验的逃避潜伏期较MECT组、丙泊酚组及抑郁组延长而空间探索时间缩短(P0.05);MECT组和ECT组较丙泊酚组、抑郁组SYP蛋白及其mRNA表达明显升高(P0.05),而MECT组较ECT组的表达减少(P0.05)。结论丙泊酚可改善抑郁大鼠电休克后学习记忆功能,其机制可能与抑制电休克后海马SYP的过度表达有关。
