高压氧对血管内皮细胞损伤大鼠纤溶系统和血小板的影响

摘    要:目的:通过观察高压氧(HBO)对血管内皮损伤大鼠纤溶系统以及血小板的数量(PLT)及另2项参数的影响,以证实HBO对血栓的发生,发展过程有否抑制作用。方法:将44只大鼠随机分成6个组,正常组(N组,n=6)、模型对照组(M组,n=10)、模型制作后HBO治疗5天组(T1组,n=7)和10天组(T2组,n=7)以及高压空气治疗5天组(T3组,n=7)和10天组(T4组,n=7)。血管内皮细胞(VEC)损伤动物模型均以皮下注射大剂量肾上腺素制成。测定其血浆中纤溶酶原激活物抑制物(PAI)和组织型纤溶酶原激活物(tPA)的活性以及PLT和另2项参数。结果:T1组和T2组比M组PAI活性明显较低,tPA活性和PLT显著较高(P<0.05,P<0.01);T3组和T4组较M组PAI、tPA活性及PLT变化不明显(P>0.05)。结论:HBO可以促进血管内皮损伤大鼠纤溶活性的恢复,减少血小板的聚集,促进内皮细胞损伤的修复,有效地抑制血栓前状态,从而预防血栓的形成。

Abstract:Objective To investigate if hyperbaric oxygen could inhibit the thrombosis by observing fibrinolysis system,platelet count(PLT)and another two parameters in SD rats with injured vascular endothelial cells(VEC).Methods 44 rats were randomly divided into 6 groups:normal group(group N, n =6);model group(group M, n =10);group T\-1( n =7)were treated by hyperbaric oxygen for 5 days after the model was made;group T\-2( n =7) were treated by hyperbaric oxygen for 10 days after the model was made;group T\-3( n =7) were treated by hyperbaric air for 5 days after the model was made;group T\-4( n =7) were treated by hyperbaric air for 10 days after the model was made.The rat model with VEC was injury established by injecting high dose of adrenaline subcutaneously.The tissue type plasminogen activator(tPA) activity,plasminogen activator inhibitor(PAI) activity,PLT and another 2 parameters were measured.Results PAI activity both in group T\-1 and in group T\-2 was significantly lower than that in group M( P <0 05,0.01).tPA activity and PLT both in group T\-1 and in group T\-2 were significantly higher than those in group M( P <0 05,0.01).PAI activity,tPA activity and PLT did not show marked changes in group T\-3 and group T\-4,in comparison with group M( P >0 05).Conclusions HBO could significantly promote the recovery of fibrinolysis activity,decrease platelet aggregation,improve the recovery of injured VEC,inhibit the prethrombotic state effectively and prevent thrombosis.
