大肠癌组织中的端粒酶活性变化

目的：研究端粒酶活性与结直肠癌恶性进展的关系。方法：应用端粒重复扩增（ＴＲＡＰ）方法检测２２例大肠腺瘤、４３例结直肠癌及邻近组织中的端粒酶活性，并分析其与临床病理的关系。结果：端粒酶活性阳性率在大肠癌中为９５．４％，腺瘤中为９．１％，而邻近组织中仅为２．３％，在低、未分化癌及粘液腺癌中端粒酶活性呈中、强表达者占多，而在中、高分化癌包括乳头状腺癌及管状腺癌中存在相当低、弱表达，但其总的表达在大肠癌Ｄｕｋｅｓ各期中无甚差异。结论：大肠癌的恶性转化与端粒酶的活化有关，检测端粒酶活性对临床大肠癌尤其是早期大肠癌具有特别重要的诊断意义。

Telomerase activity in human primary colorectal cancer

Purpose: To explore the association between telomerase activity and malignant progression of human colorectal cancers.T5HZMethods: Telomerase activity was measured in 22 colorectal adenomas,43 colorectal cancers and corresponding normal tissue using a telomeric repeat amplification protocol(TRAP),and results were compared with clinical staging data of the tumor. Results:ZTelomerase activity was present in 9.1% of the adenomas,95.4% of the tumors and only 2.3% of normal tissues(P<0.01).Telomerase activity levels were high in poorly differentiated carcinomas including mucinous adenocarcinomas while expression of the enzyme was low or weak in welldifferentiated carcinomas including papillary and tubular adenocarcinomas.In addtion,telomerase activity was also found in all stages of the tumour. Conclusion: Telomerase activity may be a universal diagnosis marker in colorectal tumor progression.
