细胞毒T淋巴细胞相关抗原4（CTLA-4）基因多态性与牙周炎患者的关系研究

目的：通过检测CTLA-4＋49A/G,-318C/T基因型在牙周炎患者中的分布进而分析CTLA-4基因多态性与牙周炎发生、发展的相关性。方法：采用多聚酶链反应-限制性内切酶片段长度多态性（PCR-RFLP）检测55例慢性牙周炎,48例侵袭性牙周炎以及80例健康对照者中CTLA-4＋49A/G,-318C/T基因型频率,统计分析其在慢性牙周炎及侵袭性牙周炎患者中的差异性。结果：CTLA-4＋49等位基因G及基因型GG在慢性牙周炎患者中的频率显著高于健康对照组,而在侵袭性牙周炎患者中无显著差异;而无论在慢性牙周炎还是侵袭性牙周炎中CTLA-4-318C/T等位基因及基因型频率均无显著差异。结论：CTLA-4基因多态性与牙周炎的发生、发展可能存在相关性,CTLA-4＋49等位基因G及基因型GG是慢性牙周炎的易感因子;此外,CTLA-4-318C/T基因多态性与牙周炎的易感性无关。

Gene polymorphism of cytotoxic T lymphocyte associated antigen--4 in Chinese periodontitis patients

Objective：To explore the potential correlation between gene polymorphism of CTLA-4 and pathogenesis of periodontitis by detecting the distribution of CTLA-4＋49A/G and -318C/T in patients with periodontitis. Methods.. The genotype frequencies of CTLA-4＋49A/G and -318C/T in 55 cases of chronic periodontitis,48 cases of aggressive periodontitis and 80 healthy volunteers were evaluated by PCR -RFLP. Results：The frequencies of allele ＂G＂ and genotype ＂GG＂ at CTLA-4＋49 site were significantly higher in patients with chronic periodontitis but not in patients with aggressive periodontitis than those in healthy volunteers. But the frequencies of allele and genotype at CTLA-4-318C/T site in patients with chronic or aggressive periodontitis were not different with those in healthy volunteers. Conclusion： Gene polymorphism of CTLA-4 may be implicated in the pathogenesis of periodontitis. The allele ＂G＂ and the genotype ＂GG＂ are the susceptible factors to chronic periodontitis. Besides, gene polymorphism of CTLA- 4-318C/T is not related with the susceptibility to periodontitis.
