低频超声联合顺铂对卵巢癌细胞影响研究

摘    要:目的探讨低频超声联合顺铂对人卵巢癌A2780细胞增殖和凋亡的影响。方法培养人卵巢癌A2780细胞,将其随机分为对照组、低频超声组、顺铂组和联合组。低频超声组给予超声840 k Hz,0.75 W/cm23 min辐照;顺铂组给予顺铂5 n M,培养48 h;联合组给予低频超声+顺铂处理。应用CCK-8检测各组细胞的增殖情况;流式细胞仪检测各组细胞凋亡以及细胞周期。结果与单独应用低频超声或顺铂比较,低频超声联合顺铂抑制人卵巢癌A2780细胞增殖的能力更强,更明显地诱导癌细胞的凋亡以及细胞周期S期阻滞,差异有统计学意义(P<0.05)。结论低频超声联合顺铂可抑制人卵巢癌细胞的增殖,诱导癌细胞的凋亡以及细胞周期S期的阻滞作用。

Impact of low-frequency ultrasound combined cisplatin on ovarian cancer cells
Abstract:Objective To investigate the effect of low-frequency ultrasound combined with cisplatin on proliferation and apoptosis of ovarian cancer A2780 cells. Methods The ovarian cancer A2780 cells were cultured,cells were randomly divided into the control group,the low-frequency ultrasound group,the cisplatin group and the combination group( low-frequency ultrasound combined cispla-tin) . The cells of low-frequency ultrasound group were given 840 kHz,0. 75 W/cm2 for 3 minutes of irradiation;cells of the cisplatin group were given concentration of 5 nM for 48 hours;cells of the combination group was given low-frequency combined cisplatin. CCK-8 assay was used to detect cell proliferation, and flow cytometry was used to detect apoptosis and cell cycle in each group. Results Compared with the effects of the low-frequency group and the cisplatin group,the effect of low-frequency ultrasound com-bined cisplatin significantly enhanced inhibiting the proliferation of A2780 cells,inducing cell apoptosis and promoting S-phase block, and the differences were statistically significant(P<0. 05). Conclusion Low-frequency ultrasound combined with cisplatin can en-hance the inhibitory effect of proliferation,induce cell apoptosis and promote S-phase block.
