黄连解毒汤对衰老小鼠学习记忆的影响

摘    要:目的:探讨黄连解毒汤对衰老小鼠的抗衰老作用及其机制。方法:采用皮下注射D- 半乳糖的方法制备衰老小鼠模型。将ICR小鼠随机分为正常对照组、模型组、1、2、4和8g/kg黄连解毒汤生药量处理组,采用自发活动实验检测小鼠的自主活动,用跳台实验检测小鼠的学习记忆能力,用硫代巴比妥酸法和黄嘌呤氧化酶法测定脑组织中超氧化物歧化酶(SOD)和丙二醛(MDA)含量。结果:与正常对照组相比,模型组小鼠的学习记忆能力减退,MDA含量增加,SOD活性降低。黄连解毒汤各剂量组小鼠的学习记忆能力明显改善,与1g/kg和8g/kg相比,2g/kg和4g/kg黄连解毒汤组小鼠脑内的MDA含量明显降低,SOD活性显著增高。结论:黄连解毒汤能显著改善口半乳糖衰老小鼠学习记忆能力,其机制可能与其抗氧化特性有关。

Abstract:To investigate the potential anti-aging effect of H uanglian-Jie-Du-Tang (HJDT) and explore the mechanism in mice. METHODS: The ICR mice were divided into normal control group, aging model group and HJDT treated group (1, 2, 4, and 8 g/kg). Ag ing mice were induced by subcutaneously injec tion with D-galactose. The spontaneous activity and the learning and memory ability were evalu ated by a locomotor test and step down test. The SOD activity and the MDA level were meas ured by xanthine oxidase method and thiobarbi turic acid method. RESULTS: Compared withthe normal control group, the learning and memory ability and the MDA level decreased and the SOD activity increased in aging mice, which were markedly reversed by HJDT (2 and 4 g/ kg). But there was no significant effect of HJDT (1 and 8 g/kg). CONCLUSION. HJDT amelio rates learning and memory impairment in D-ga lactose-induced aging mice through its potent an tioxidant activities.
