cAMP-PKA-CREB信号通路在大鼠酒精性肝纤维化星状细胞模型中的作用

目的探讨cAMP-PKA-CREB信号通路在大鼠酒精性肝纤维化星状细胞模型中的作用。方法采用不同浓度乙醛不同时间间隔干预肝星状细胞(HSC),建立体外酒精性肝纤维化星状细胞模型;采用MTT法检测HSC增殖,确定造模浓度及时间;RT-PCR法检测HSC活化指标α-SMA mRNA表达情况;125I-cAMP放射免疫分析方法测定正常组与模型组HSC内cAMP含量;RT-PCR法测定PKA,CREB mRNA表达情况。结果采用浓度为200μmol.L-1乙醛刺激HSC 48 h可建立酒精性肝纤维化星状细胞模型;模型组α-SMA mRNA表达较正常组明显增强,模型组cAMP含量较正常组显著增加;PKA,CREB mRNA表达也较正常组明显增强。结论乙醛可刺激HSC增殖活化,其机制可能与其激活HSC cAMP-PKA-CREB信号通路有关。

Effect of cAMP-PKA-CREB signal pathway in the model of alcoholic hepatic fibrosis stellate cells isolated from rats

Objective To explore the effect of cAMP-PKA-CREB signal pathway in the model of alcoholic hepatic fibrosis stellate cells from rats. Methods We detect the absorbance of 25,50,100,200,400 μmol · L^-1 acetaldehyde to HSCs for 24,48,72 hours to estab- lish the model,the mRNA expression of α-SMA,the activation index of HSCs,was detected by RT-PCR,the cAMP content was detected by 125 I-cAMP radioimmunoassay, PKA and CREB were measured by RT-PCR. Results We establish alcoholic liver fibrosis modelwith acetaldehyde（200 μmol · L^- 1） for 48 hours,The mRNA expression of a-SMA in the model was more than normal,and the cAMP in the model was more than the normal. The mRNA expressi., of PKA and CREB were also more than normal. Conclusion The acetaldehyde can stimulate HSCs , and promote the HSCs to proliferate and active. The mechanism may be involved with acetaldehyde activates the eAMP-PKA-CREB signal pathway in HSCs.
