高良姜防治糖尿病肢端疼痛的潜在靶点及作用研究

摘    要:目的用网络药理学的方法探讨高良姜治疗糖尿病肢端疼痛的分子作用机制。方法通过中药系统药理学数据库与分析平台筛选高良姜的有效成分,用药物银行数据库、基因卡片数据库预测和筛选高良姜有效成分和糖尿病肢端疼痛相关靶点,用String数据库绘制靶点相互作用网络,借助生物学信息注释数据库对靶点进行基因本体和基于京都基因与基因组百科全书(KEGG)分析,用KEGG数据库构建高良姜治疗糖尿病肢端疼痛的通路图。结果筛选得到高良姜13个有效成分,涉及88个靶点。靶点相互作用网络结果表明,高良姜的靶点主要涉及氧化应激、炎症和细胞凋亡等生物过程,通过调节丝裂原活化蛋白激酶、肿瘤抑制基因(p53)和核因子κB等信号通路来发挥治疗糖尿病肢端疼痛的作用。结论高良姜通过多成分-多靶点-多通路协同治疗糖尿病肢端疼痛,其主要作用机制可能涉及抗氧化、抗炎和抑制细胞凋亡。

Abstract:Objective To explore the molecular mechanism of Alpiniae Officinarum Hance in the treatment of diabetic extremity pain by the network pharmacology. Methods The active ingredients of Alpiniae Officinarum Hance were obtained through traditional Chinese medicine systems pharmacology. Drugbank and GeneCards databases were used to predict and screen the related targets of Alpiniae Officinarum Hance and diabetic extremity pain. The protein interaction network was constructed using the String database. The gene ontology and Kyoto encyclopedia of genes and genomes( KEGG) pathways involved in the targets were analyzed by the database for annotation,visualization and integrated discovery. The KEGG database was used to construct a pathway map of Alpiniae Officinarum Hance in the treatment of diabetic extremity pain.Results The results showed that 13 active components and 88 targets of Alpiniae Officinarum Hance were involved. The results of the target interaction network showed that the biological process of oxidative stress,inflammation and apoptosis were mainly involved,which played a role in the treatment of diabetic extremity pain by regulating the Mitogen-activated protein kinase,tumor suppressor gene( p53),nuclear factor kappa-B and other signal pathways. Conclusion Alpiniae Officinarum Hance treats diabetic extremity pain through multiple components,multiple targets,multiple pathways,its mainly mechanism of action involved antioxidation,anti-inflammatory and anti-apoptosis.
