肾上腺髓质素基因敲除小鼠单侧输尿管结扎肾脏α-SMA表达特点及缬沙坦的作用

摘    要:目的观察肾上腺髓质素基因缺失状态下小鼠肾脏α-SMA的表达特点及缬沙坦的作用。方法结扎单侧输尿管方法制备肾间质纤维化实验动物模型,采用免疫组化、RT-PCR方法检测α-SMA的定位与mRNA表达。结果单侧输尿管结扎小鼠肾脏α-SMA表达明显高于假手术组(P<0.01),其中肾上腺髓质素基因敲除小鼠高于野生小鼠,缬沙坦可下调其表达。结论单侧输尿管结扎可诱导肾小管上皮细胞表型转化,肾上腺髓质素基因缺失状态下其表达增强,血管紧张素受体拮抗剂缬沙坦可部分抑制其转化。

Abstract:Aim To observe the expression ofα-smoothmuscle actin(α-SMA) in adrenomedullin knockoutmice with unilateral ureteral obstruction(UUO) andthe effectofvalsartan.Methods Renal interstitial fi-brosis animals were made with UUO、α-SMA and theexpression of mRNA were examined in adrnomedullinknockout(AMKO) mice and wild type(WT) mice byimmunohistochemistry and RT-PCR.Results The ex-pression ofα-SMA in UUO groups were up-regulatedsignificantly compared with sham group(P<0.01),specically in AMKO group.Valsartan has the effect indown-regulating its over-expression.Conclusion Tu-bular epithelial-myofibroblast transdifferentiation wasinduced by UUO and over-expression in AMKO mice.Valsatan has a role to inhibit it partly.
