摘    要:目的观察肉桂酸对胃腺癌MGC-803细胞的诱导分化作用。方法肉桂酸(1、2和3mmol.L-1)作用于MGC-803细胞后,应用四甲基偶氮唑蓝(MTT)法测定细胞增殖能力、平板克隆试验测定细胞集落形成率、流式细胞仪测定细胞周期、TRAP-银染法测定端粒酶活性。结果肉桂酸作用24、48、72和96h后胃腺癌细胞生长均明显受抑制,作用48h后细胞周期出现向G0/G1期移行的特征性动力学改变。端粒酶活性明显受到抑制。细胞集落形成率明显下降(P<0.05)。结论肉桂酸对胃腺癌细胞有诱导分化作用。

Abstract:Aim To investigate effects of cinnamic acid (CINN) on the differentiation of MGC-803 cells. Methods MGC-803 cells were treated by CINN (1, 2 and 3 mmol·L-1), and the rate of growth suppression was evaluated by 3-(4,5-dimethylthiazole-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. Plate clone formation assay was carried out to detect the phenotypes of colony formation. FACS analysis was used to analyze the cell cycle. The activity of telomerase was examined by telomeric repeat amplification protocol (TRAP) PCR-silver staining. Results CINN showed significant inhibitory effect on growth of these gastric carcinoma cells at 24, 48, 72 and 96 h. Treated with CINN for 48 h, cell cycle showed G0/G1 phase arrest, and the activity of telomerase was inhibited significantly. The colony-forming rate was reduced drastically (P<0.05). Conclusion CINN induces differentiation of MGC-803 cells.
