胆碱能药物和人眼组织的M_3受体

目的　检测M3受体在人眼组织中的分布 ,探讨M3受体和胆碱能药物的关系。方法　人工合成M3受体特异性胞外肽链段 ,采用杂交瘤技术 ,制备M3受体单克隆抗体 ;用ELISA试验、放射配基受体结合实验和免疫组织化学实验 ,鉴定该抗体的特异性 ;通过免疫组织化学技术 ,检测M3受体在人眼组织中的分布。结果　虹膜、睫状肌、睫状体非色素上皮、视网膜和小梁网组织分布有M3受体 ,其中睫状肌的纵行纤维的M3受体的分布明显多于环行纤维。结论　人眼组织中广泛分布M3受体 ,是胆碱能药物主要作用的受体

Cholinergic drugs and M3 receptor in human eye

AIM To measure distribution of M 3 receptor in the human eye; to explore the relationship between M 3 receptor and cholinonergic drugs. METHODS Peptides of human M 3 receptor in extracellular amino terminal were chemically synthesized,monoclonal antibody was prepared by hybrido ma technology. Specificity of antibody was tested by enzyme linked immunosorbent assay, immunohistochemistry, and radioligand binding assay of M 3 receptors. The distribution of M 3 receptor in the human eye studied with immunohistochemistry was tested by using monoclonal antibody. RESULTS The results indicate the antibody has highly specificity. M 3 receptors exist in ciliary muscle, iris, nonpigmented epithelium of ciliary body, trabecular meshwork and retina. The M 3 receptors in the vertical of ciliary muscle are more than that in the longitunal of ciliary's muscle. CONCLUSION M 3 receptor exists extensively in human eye.It is the binding receptor of cholinergic drugs.
